| Purpose: Effectofelectroacupuntureonchronicunpredictablestimulationmethod, combined with subcutaneous injection of carbon tetrachloride solutionnatural perimenopausal rats was established in perimenopausal depression hepatic depression syndrome model rats estradiol (E2) levels, as well as5-serotonin2A (5-HT2AR) and extracellular regulated protein kinase (ERK1/2) expression;to electroacupuncture treatment of perimenopausal depression effects and mechanisms of action.Method:Select73SPF grade11-13months natural perimenopausal female SD ratas experimental object,the weight is260-471g, were randomly divided into modelgroup19, traditional Chinese medicine group18, western medicine group, electro-acupuncture group18, in addition to select20SPF grade3-4months old female SDrats as youth group for140-191g, body weight. In addition to the young group,the remaining four group by chronic unpredictable stimulation model, at the sametime, abdominal subcutaneous injection of4%carbon tetrachloride peanut oilsolution made the hepatic depression syndrome. Treatment means respectively,Chinese medicine group gavage Xiaoyao Pill in water solution (0.2g/100g/D);western medicine group gavage clomipramine gavage (3mg/100g/D); electro-acupuncture group were trained, Sanyinjiao, Zusanli, liver Yu, kidney andacupoint therapy; three groups treated continuously for10days. Model group weregiven saline3ml/day, the youth group of the corresponding sham stimulation.In experiment one day before and twenty-eighth day experiment respectivelyweighing each rat body weight and open field test; twenty-ninth days weresacrificed rats in each group, through the abdominal aorta blood and ice removedcraniotomy in hippocampus brain tissue,70℃environmental conservation. ELISAmethod for the detection of rat serum estradiol concentration(E2), RT-PCR methodfor the detection of rat brain hippocampal tissue5HT2A (5-HT2A) receptor andimmunohistochemistry rat brain hippocampus extracellular protein kinase (ERK1 /2) and related results.Results:1. Model group and young group comparison, E2detection values were significantlyreduced (P <0.01). After the treatment of traditional Chinese medicine,acupuncture group and Western medicine group, group E has improved, the threegroups compared with the model group had statistical significance (P <0.05).But the electroacupuncture group and Western medicine group, Chinese medicinegroup had no statistical significance (P>0.05).2.Studies show that rat brain hippocampal5-HT2A receptor expression level inmodel group were increased, model group and young group comparison of rat brainhippocampal5-HT2A receptor expression levels significantly increased withsignificant difference. Compared with the model group in each treat ment groupand acupuncture group and Western medicine group, Chinese medicine group wereimproved, perimenopausal depression rat brain hippocampal5-HT2A receptorexpression levels were higher in the state.3. Model group in the rat hippocampal ERK1/2expression was significantly lower,model group and young group expression have the obvious difference(P <0.01);electro acupuncture group, western medicine group, Chinese medicine group andmodel group than in the ERK1/2expression improves, with statistical significance(P <0.01). Electric acupuncture group and Western medicine group, and Chinesemedicine group, three groups showed no statistical significance(P﹥0.05).Conclusion:1.EA can effectively regulate the levels of E2, on the aging rats serum E2regulation has a certain effct.2. Effects of Electroacupuncture on brain and hippocampal5-HT2A receptorexpression has a negative regulatory role, has a role in the treatment ofdepression.3.Electric brain response to hippocampal ERK1/2expression with positiveregulation effect on depression, therapeutic effect. |
