Objective To observe the effect of herbal-cake-separated moxibustion on chronic bronchitis(CB) model rats in broncho-alveolar lavage fluid (BALF) neutrophil count, lung tissue intercellular adhesion molecule-1(ICAM-1) mRNA, protein, p38MAPK signal transduction pathway activation marker phosphorylated p38MAPK (p-p38MAPK) protein.discussing the immune regulating mechanism of cake-separated moxibustion on CB model rats based on p38MAPK signal transduction pathway.Methods Using the modified fumigation way to copy the CB model rats,48SD rats were paired before randomly divided into four groups: blank group, model group, herbal-cake-separated moxibustion group, paperwasher-separated moxibustion. Before and after the treatment drawed materials in twice, take herbal-cake-separated moxibustion as subjects factor, testing the neutrophil count in BALF, the mRNA and protein of ICAM-1, the protein of p-p38MAPK in lung tissue. Compare the differences in and between groups before and after treatment.Results (1) the herbal-cake-separated moxibustion and paperwasher-separated moxibustion can reduce the total number of cells (P<0.05) and neutrophil count (P<0.05) in BALF, and the herbal-cake-separated moxibustion is more effectively (P<0.05).(2) the herbal-cake-separated moxibustion and paperwasher-separated moxibustion can reduce ICAM-1mRNA and protein in lung tissue(P<0.01, P<0.05)and the herbal-cake-separated moxibustion is more effectively (P<0.05).(3) the herbal-cake-separated moxibustion and paperwasher-separated moxibustion can reduce p-p38MAPK protein in lung tissue(P<0.01)and the herbal-cake-separated moxibustion is more effectively (P<0.05).(4) the pathological tissue of rat lens improved fumigation method after modeling the observed:alveolar expansion, infiltration of inflammatory cells, tracheal epithelial cell hyperplasia, local epithelial cell loss, shorter cilia sparse and even local shedding, within the lumen of many inflammatory secretions, mucous membrane lower inflammatory cell infiltration. After the treatment, herbal cake-separated moxibustion group: structure of basic integrity, no tracheal mucosa epithelial cell shedding, tube wall has mild hyperemia and edema, cilia, but normal distribution density and length were worse than the blank group, infiltration of inflammatory cells reduced obviously;moxibustion group:tracheal mucosa structure fuzzy boundaries, the pipe wall with mild hyperemia and edema cilia, with sparse, distribution is not uniform, but compared with the model group, length and density was high, with a small amount of inflammatory cell infiltration. Conclusion (1) herbal-cake-separated moxibustion can reduce the total number of cells and neutrophil count,prevent inflammation from developing (2) herbal-cake-separated moxibustion can inhibit the endothelial cells from producing ICAM-1, stop the aggregation of neutrophil, thereby modulating the inflammatory response;(3) herbal-cake-separated moxibustion could regulate endothelial cells, neutrophils and treat chronic bronchitis through p38MAPK signal pathway;(4) herbal-cake-separated moxibustion and paperwasher-separated moxibustion both are effective treatment for chronic bronchitis model rats, but the effect of herbal-cake-separated moxibustion plus paperwasher-separated moxibustion.
