ObjectiveThe experiment takes spontaneously hypertensive rats (Hypertension Rat, SHR) as models, with quantitative PCR as technology. It aims at exploring twirling reinforcing-reducing techniques on the effects of differences by observing the blood pressure of spontaneously hypertensive rats and hypothalamus in the TGF-β of mRNA changes in p38protein kinase mRNA expression after applying the twirling reinforcing-reducing techniques on Taichong (LV3).Method75clean grade, malespontaneously hypertensive rats (Hypertension Rat, SHR),;15male Wistar-Kyoto (WKY) rats of clean grade. The former were randomly divided into five groups: reducing with twisting and twirling group, reinforcing with twisting and twirling group, acupuncture control group, Taichong (LV3) pressing group and model control group; WKY rats are treated as blank controlgroups. Rats in each group in the animal laboratory, feeding14days to adapt to the environmentRats’initial blood pressure was measured at the very beginning of the experiment. Formal experiment in rats begun after adaptation to the environment. For reducing with twisting and twirling group and reinforcing with twisting and twirling group, the needle manipulation was applied on Taichong (LV3). acupuncture control group only needling bilateral Taichong (LV3) with no manipulation; the Taichong-pressing group received pressing bilateral Taichong (LV3) with needle-handle as intervention. The model control group and blank control group received no stimulation excepting binding. It takes1minute for twisting and twirling and a20minutes needling retention, once every day,14days as treatment. After the end of the last acupuncture treatment, measuring blood pressure. The rats were decapitated, craniotomy to remove the hypothalamus for detecting the expression of TGF-β mRNA in the hypothalamus with Q-PCR.Result1changes in blood pressureBefore the acupuncture treatment, there’s no no significant difference (P>0.05) on each group of SHR’s systolic blood pressure. Compared with WKY rats group, SHR rats’s systolic blood pressure was significantly high (P<0.01). After14days of acupuncture treatment, all groups except the reducing with twisting and twirling group of SHR rats’s systolic blood pressure have increased to varying degrees. Statistical analysis showed a recreased systolic blood pressure in reducing method group, and the result has significant difference with the reinforcing group (P<0.05), and with other SHR groups (P<0.01). It has no significant difference (P>0.05) with that before the treatment. There’s significant difference between low systolic blood pressure of the reinforcing group and all other groups (P<0.05), but no significant difference with that of the acupuncture control group (P>0.05) and before the acupuncture treatment (P>0.05); The acupuncture control group has a significant difference with two twisting and twirling groups and the control group(P<0.01) and systolic blood pressure increased in twisting reducing method group, but with the combination of twisting reinforcing law non-acupuncture group compared with no significant difference (P>0.05); the Tai Chong press group and model control group systolic blood pressure higher than before the acupuncture treatment, with a significant difference (P<0.05); blank control group, systolic blood pressure was increased, but not statistically significant (P>0.05).2TGF-β mRNA expression in the hypothalamusThere was a significant difference (p<0.01) between reducing with twisting and twirling group and model control groupand acupuncture control group (p<0.05); the reinforcing group, the Taichong pressing group, and the blank blank control group has no significant difference with that of the reducing grcup(P>0.05). There’s no significant difference (P>0.05) between the reinforcing group and acupuncture control group, Taichong pressing group, model control group, and the blank control group. There’s no significant difference between acupuncture control group and Taichong (LV3) pressing group, the model control group, and the blank control group (P>0.05). Taichcng press group and model control group, the control group showed no significant differences (P>0.05). Model control group and the control group were significantly different (p<0.05).3p38mRNA expression in the hypothalamusThere was a significant difference between the reducing group and the acupuncture control group (p<0.01); and a significant difference (p<0.05) with that of the reducing group and model control group. It has no significant difference with Taichong pressing group and the blank control group (p>0.05).There was a significant difference (p<0.01) bewteen reinforcing group and the acupuncture control group; a significant difference (p<0.05) between reinforcing group with Taichong pressing group, and no significant difference between reinforcing group with the model group, the blank control group (p>0.05).Compared with the Taichong pressing group, model control group, and the bland control group, acupuncture control group has a significant difference (p<0.01).Taichong pressing group and model control group has a significant difference (p<0.05); but there’s no significant difference (p>0.05) compared with the control group.Model control group and the control group has a significant difference (p<0.05).Conclusion1The acupuncture Taichong(LV3) twirling reinforcing-reducing techniques can effectively reduce systolic blood pressure of SHR rats, there are difference in the effect on the presence,with reducing method better than twisting reinforcing.2About the regulation of blood pressure by twisting and twirling methods, there’s a relation between reducing method and TGF-β of mRNA, p38protein kinase mRNA levels. Twisting reinforcing method of blood pressure regulation and TGF-β of mRNA, mRNA levels of the p38protein kinase relationship is not obvious.
