From The Perspective Of Oxidative Stress Of Qi And Activating Blood On Diabetic Nephropathy Proteinuria

Propose:Diabetic nephropathy has become a worldwide public health problem which is a serious harm to human health.Protein urine is one of the main clinical manifestations of DN, an independent risk factors for DN,and an important clinical index of disease progression and DN forecasting long-term prognosis.Strict control of urinary protein has become one of the foundation treatment to delay the progress of DN.The domestic and foreign scholars have devoted to the control of DN patients with proteinuria to delay DN,and made some progress. The study on clinical effect of Yiqi Yangyin Huoxue Recipe effect on proteinuria in DN rats and the mechanism of oxidative stress.Methods:This experiment used90healthy male Wistar rats (clean SPF), about180-200g, after the adaptability fed for1weeks, the healthy, good condition, and normal blood glucose, no urine protein rats are choosed to be observed. The rats were randomly divided into normal control group and model group. Normal control group need not operation treatment, model group, need to do the left kidney ligation operation. After2weeks, rats in normal group were fed with normal diet, model group by feeding high fat food. After4weeks, all groups of Wistar rats fasted for12hours, the module need a single intraperitoneal injection of streptozotocin (STZ)30mg/kg, and the normal control group rats were intraperitoneally injected with equal volume of the buffer solution. After72h, blood glucose of tail vein was higher than that in16.7mmol/L; urine volume> urine volume150%; urinary protein excretion of urinary>30mg/24h were choosed as modeling success,and included in the object of observation. The model rats were numbered and randomized diabetic nephropathy, model group, Yiqi Yangyin Huoxue Recipe of high, medium and low dose group, Chinese medicine Tangmaikang group, with15rats in each group. Normal group were8rats. Rats in normal group were fed with normal diet, the other groups were fed with high energy diet. According to body weight were fed with drugs with the same volume. After administration of0,2,4weeks, blood glucose of tail vein was measured, at the same time, moving the rats to the metabolic cage, collecting24h urine. On the4weekend,after the rats anesthesia,blood was taken from the abdominal aorta, serum after centrifugation, do the biochemical test by automatic biochemic alanalyzer, at the same time, taking the right kidney, and incision the right kidney along the longitudinal, half was fixed in10%formalin, pathologic HE and Masson staining, observed under light microscope, the other half and stored in liquid nitrogen, protein of kidney tissue was detected by coomassie brilliant blue method,glutathione protein carbonyl content, renal tissue detection of peroxidase (GSH-px) activity, malondialdehyde content and succinate dehydrogenase, catalase, superoxide dismutase, testing content and mitochondrial membrane potential of mitochondrial ROS in renal tissue. In order to reveal the effect of Yiqi Yangyin Huoxue Recipe on DN and the mechanism of oxidative stress. Results:1、Yiqi Yangyin Huoxue Recipe on diabetic nephropathy rats spirit, activity, hair color and general state has certain improving effect, and can regulate blood lipid abnormality in a certain extent.2、Yiqi Yangyin Huoxue recipe can improve diabetic nephropathy rats to a certain extent,24hours urine protein.3. Yiqi Yangyin Huoxue recipe can reduce glomerular basement membrane in DN rats, improve mesangial DN rat broadening and mesangial matrix accumulation.4. Yiqi Yangyin Huoxue recipe can obviously inhibit the diabetic nephropathy rats kidney protein carbonyl content and increase GSH-PX activity, and renal tissue SOD, SDH and MDA and the model group showed no significant difference.5、Yiqi Yangyin Huoxue Recipe group kidney mitochondrial RFU values were higher than that in the model group, Yiqi Yangyin Huoxue Recipe group kidney mitochondrial ROS were higher than the model group decreased, with statistical significance (p<0.05).Conclusion:Yiqi Yangyin Huoxue decoction has obvious effect on the general state of diabetic nephropathy rats, and can regulate the blood lipid abnormality in a certain extent, a reduction of24hours urinary protein excretion, renal function, renal pathological changes, can enhance the activity of DN in rat GSH-PX, improving renal mitochondrial RFU and mitochondrial ROS.
