As a model plant, Arabidopsis thaliana is the first higher plant of which the genome sequencing has been accomplished.Further research has been shedding light on functions of numerous genes, however, there are still piles of unidentified genes to be studied. The study on localization, function or expression profile and regulation of these genes interests people a lot along with the improvement made on bioinformatics, reverse genetics and mordern molecular biology. In this research, we isolated a new gene overlapped in2SSH libraries from rapeseed of different stages and cDNA library and nominated it as BnaCS25, followed by bioinformatical analysis, we found another new gene in Arabidopsis, AtCS25the homolog of BnaCS25and it may be invovled in pollen or cell wall development. In this research, AtCS25was preliminarily studied based on molecular biology and bioinformatics methodology and there are some findings.1.Bioinformatics analysis indicates AtCS25is plant-exclusive and was located in A. thaliana choromosome2.It contains2exons and2introns respectively,with752bp full length CDS,it encodes a protein of184aa with a MW of19KD.The protein contains a pollen-allergic domain PollenOlee1/extensin with transmembrane stucture and is probablely to be a secretion protein.2.The exprssion profile of AtCS25was studied using RT-PCR,the results showed its expression level was higher in only stage of development while detectable in roots in mature plants.3.This reserch cloned the AtCS25and constructed the O-E-V pBI121:AtCS25and RNAi vector pGSA1285:AtCS254.A.tumefaciens mediated transformation of O-E-V was performed using floral-dipping method in Col-0background. This reserch obtained35transgenic lines and RT-PCR anlysis indictated AtCS25was overexpressed in transgenic plants.
