In normal physiological processes and follicle development process, the granulosa cellsdivided into mural granulosa cells (mGCs) and cumulus cells (CCs), respectively expressingtheir own special subset of transcription. Both of the two have a clear division of labour: CCsaffect oocyte maturation, and mGCs main endocrine function. COCs, isolated from thefollicles, are cultured in vitro, COCs and mGCs secretion factors are separated. In the processof oocyte in vivo maturation, granulosa cells is the main functioning cell of ovarian,itsproliferation and differentiation directly affects the launch of ovarian follicles growth,development, ovulation, corpus luteum formation and ovarian function activities such assteroid hormone secretion. Oocyte (OC) guides the granule cells(GCs) proliferation anddifferentiation; simultaneously GC affect the oocytes maturation. There are many factorsinvolved in the regulation of the process. Cumulus cells encircle around oocyte, clinging tothe closest relations with oocytes, in the process of oocytes IVM, nutrients in the culture fluidthrough the connection between granular cell and oocytes into oocytes, in order to promotingoocytes maturation, in the process, cumulus cells occur a phenomena of expansion, thatcumulus expansion and oocyte maturation have a certain extent relationship, cumulus cellsexpansion is mainly due to the genes expressions of cumulus cells, and oocytes may also playa role in the process, cumulus cells provide some secretion factors for oocytes maturation toaffect and stimulus maturation, in turn, oocytes after matured can secrete some substance tostimulate cumulus expansion.The purpose of this study is that:(1) detection the genes expressions of cumulus cellsexpansion;(2) the influence of GSK3inhibitors to oocyte maturation and cumulus cellsproliferation;(3) the cffect of GSK3inhibitors on cumulus cells apoptosis.Results:(1) In the proliferation of cumulus cells, genes related expansion such as GDF9,HAS2, PTGS2, PTX3and Tnfaip6are expressed;(2) During oocytes maturation with GSK3inhibitors, GSK3inhibitor has a certain promoting effect on oocyte maturation, but notsignificantly; and promote cumulus cells expansion, under normal circumstances, cattleoocytes maturation culture prepared with BSA cultivate oocytes, cumulus cells don’t occurexpansion, but after joining GSK3universal inhibitors, cumulus cells diffuse, oocytesmaturation is similar to the above.(3) after oocyte maturation, the degranulation of oocytes, the granulosa cells alone take train, divided into two group A/B, GSK3inhibitors were addedto the group A, group B as control group, the results found that at24h cell growth when thetwo groups is similar, but develop to48h or longer, it found that join inhibitor the growthcondition of group A was better than control group(.4)GSK3inhibitors have a certain degreeof slow effect on apoptosis of cumulus cells.Conclusion: granular cell growth, proliferation, apoptosis is associated with GSK3gene,because of GSK3as Wnt signaling pathway is one of the most important nodes, so the GSK3inhibition of Wnt pathways, in this case, the Wnt signaling pathway are suppressed, cumuluscells is simple but didn’t happen proliferation, but BSA culture diffusion occurs after joiningGSK3inhibitors, and in under the condition of the serum culture diffusion, shows that thereare similar GSK3inhibitor substances in serum. Join GSK3inhibitors cultivation system, theoocyte in the middle of the first polar body a certain discharge rate rise, prove that undernormal circumstances, after inhibition of GSK3GSK3inhibitors, activation of Wnt pathways,stable bonus on oocyte maturation. Granulosa cells by joining GSK3inhibitors in theexpression of Bax and Bcl-2gene quantity decreased, even inhibits the Bcl-2geneexpression, but look from both drop, and promote apoptosis gene Bax fall is relatively larger,showed the inhibitors to promote apoptosis gene Bax inhibitory effect more obvious,inhibitors of granulosa cell apoptosis inhibition. So the effect of Wnt signal (GSK3inhibitors)on granulosa cell is promoting proliferation, remit oapoptosis, in turn, It can be summarizedthat GSK3can inhibit the cell growth, proliferation, but a certain degree of promoting roles inapoptosis.
