At present, scientists pay more and more attention to the reservoir microorganisms, especially the16s rDNA clone library construction. This paper studies the microbial community of the high temperature and salinity oilfield formation water. Jianghan Oilfield and Shengli Oilfield formation water has a higher temperature and salt content. Special Jianghan Oilfield, the formation water temperatures up to70℃, the salt content of up to30%. This paper mainly studies the Jianghan Oilfield Guanghua and Wangchang two areas, and a few mouthfuls of Shengli Oilfield Qinghe representative wells. Analysis the reservoir microbial community in these areas, build the bacterial clone library of Jianghan Guanghua Wangchang and Qinghe region, and the positive bacteria clone library of Jianghan Guanghua and Wangchang. Screened the heavy oil activator able to activate the Qinghe and Henan Nanyang.The obtained results are as follows:(1) Jianghan Guanghua, Wangchang bacterial clone library results show that:the Jianghan Guanghua, Wangchang content a variety of oil production functional bacteria, which contains a lot of halophilic bacteria, these halophilic bacteria provide a theoretical foundation and guiding basis to carry out microbial enhanced oil recovery in high temperature and salinity reservoir. Marinobacter hydrocarbonoclasticus should be able to function as a high-salt oilfield surface wastewater treatment bacteria. In Jianghan Guanghua area has a higher levels of oil production harmful bacteria, include sulfate reducing bacteria, sulfur-oxidizing bacteria and Deferribacter thermophiles, shall take the necessary measures to prevent and treat it. In Wangchang area, Bacillus and pseudomonas in high content and rich composed, which has a high microbial enhanced oil recovery research value.(2) The clone library results of Qinghe area show that:Pseudomonasthe commom exist in three samples of the Qinghe area, mainly of them are Pseudomonas aeruginosa bacteria witch known have an oil recovery function. Bacillus in Qinghe area also may have the potential for oil recovery function. In Qinghe area there are a variety of thermophilic bacteria, witch can provide a theoretical basis and guidance to microbial enhanced oil recovery based on high-temperature reservoir. Sulfate reducing bacteria class and contents differ significantly between wells.(3) The Jianghan samples bacteria clone library results showed that:Jianghan positive bacteria constitute a relatively simple, mainly Bacillus subtilis.(4) activator screening results show that:Qinghe M137-7-X15sample activated formula:3%molasses+0.3%corn steep liquor+0.7%NH4Cl+NaNO30.3%+sodium tripolyphosphate2%+trace elements+0.2%JHME+0.05%xanthan gum.Henan Nanyang heavy sample the best activation formula:1.2%com steep liquor+0.3%NaNO3+CH3CO2Na0.3%+sodium tripolyphosphateO.1%+trace elements+0.1%JHAR+0.2%JHME+5%Rhamnolipid fermentation broth.
