Minor ginsenoside Rg3was environment-friendly prepared by conversion of materialprotopanaxadiol type saponins (PPD) using biomass material lemon as catalyst in this paper,the method analyzing ginsenosides by HPLC was founded, which provided its basis.PPD saponins were separated with D101C macroporous adsorption resin and solutionmethod. The optimum separation conditions were obtained by one-factor experiment. Theexperimential results showed that ginseng root saponins (15mg/mL) were absorbed byD101C macroporous adsorption resin at55℃for12h, desorbed by35%and80%ethanol atroom temperature for12h respectively. The purity of PPD saponins could reach98.52%byusing the solution method.The conditions of preparation of ginsenoside Rg3were optimized by one-factorexperiment and response surface methodology. The experimential results showed that itshighest yield was75.57%when PPD saponins concentration was23.64mg/mL, theconcentration of Lemon juice was97.6%, reaction temperature was85.7℃and reaction timewas130.0min.20(S)-ginsenoside Rg3,20(R)-ginsenoside Rg3, byproduct ginsenoside Rk1and Rg5were separated by the chemicial method of acetylation and deacetylation and semi-preparationHPLC, identified by HPLC, MS and NMR.The method was founded to analyze and detect ginsenoside Rb1, Rb2, Rc, Rd, Re, Rg1,20(S)-Rg3and20(R)-Rg3, which provided the basis for the study.This method is simple, environment-friendly and high-yield, which is very important tostudy the pharmacological activities of ginsenoside Rg3and develop health-care products.
