Molecular imprinting is a highly efficient technology, which can be used toprepare the high specificity selective polymers.At present, the preparation ofmolecularly imprinted polymer is mostly in the weak polar solvent, when prepare it inthe strong polar solvent, such as the system of water and alcohol, it would have poorrecognition with template molecule. It will largely limit it using in biologicalseparation. However, metal coordination effect is higher than hydrogen bondingstrength and Van der Waals force, and can exist in the system of polarity, which makethe preparation of imprinted polymer become possible in the system of water andalcohol, and has potential、broad prospects. Luteolin is a kind of important flavonoidscompounds, and it has many pharmacological activity, such as antibacterial,antioxidant, antiage and can enhance immunity.The luteolin’s metal-complexingimprinted polymer will be prepared in strong polar solution by using metalcoordination,and it is used to extract and separate luteolin, will have a broad marketand development.1. Through the molecular imprinting technique and bulk polymerization, put thecomplexes of luteolin and copper ions,functional monomer4-vinyl pyridine intomethanol/tetrahydrofuran (volume ratio,3:1) mixture solution, then add cross-linkingagent ethylene glycol dimethacrylate, initiator azodiisobutyronitrile to preparemolecularly imprinted polymer of Luteolin-Cu(Ⅱ) coordination. The process ofpreparation was optimized, and obtained the best proportion of n (Template molecule):n (Functional monomer):(cross-linking agent) was1:4:30.Based on this, The adsorptivity of the coordination and molecularly imprintedpolymer with zwitterions was discussed. After the experiment, it was found thatcoordination of molecularly imprinted polymer had the biggest adsorption, and addinga small amount of copper ions could increase the adsorptivity. The equilibriumadsorption、 adsorption dynamics and adsorption selectivity of luteolin-Cu(Ⅱ)coordination molecularly imprinted polymer was studied. The results showed that theadsorptivity of luteolin-Cu(Ⅱ) coordination molecularly imprinted polymer wasbetter than luteolin molecularly imprinted polymer.2. Luteolin-Cu(Ⅱ) coordination molecularly imprinted polymer was used as thestationary phase of the solid phase extraction column,and it was used to separateluteolin from luteolin-apigenin mixture solution.Firstly, the solid phase extraction column was washed by40%acetitrile solution, then was eluted byethanol-water(volume ratio,5:5) mixture solution.The purity of luteolin reached91.32%,and the recovery rate reached86.37%. First,5mL40%of acetonitrilesolution was used to wash the solid phase extraction column,and thenethanol-water(volume ratio,5:5) mixture solution was used to elute the solid phaseextraction,the purity of luteolin increased from2.62%to92.67%after solid phaseextraction.
