This paper selected several multi-fused ring aromatic compounds, such asFluorescein derivants (Fluorescein Fluorescein-DA and Fluorescein-DA-Fe), theCresol Red derivants (Cresol Red, Cresol Red-DA and Cresol Red-DA-Fe) andThymol Blue derivants (Thymol Blue, Thymol Blue-DA and Thymol Blue-DA-Fe).Bovine serum albumin (BSA) was used as a targeted proteins. In this thesis, it mainlyresearches whether it exist interaction between these polycyclic compounds and BSA.And then the damage effect and the injury mechanism of these three polycycliccompounds to BSA under ultrasonic irradiation were investigated. BSA is a kind ofprotein which was the important material composing biological cells, after the injuryof BSA, cancer cells can natural apoptosis, thereby inhibiting and treatmenting cancer.The cells were damaged by the reactive oxygen species and free radicals generated bysonosensitizer drugs under ultrasonic irradiation. Therefore, this paper also examinesthe species of the generation reactive oxygen species (ROS) and useOxidation-Extraction Spectrometry method to identify ROS category. This article isbeing based on the studies of activated anthraquinone derivants under ultrasound andfinding out the polycyclic structure compounds for the further experiment. Adding theamino acids and Ferric (III) to the three types of derivants, the new derivants havelaid a foundation for future clinical treatment of cancer cells. Specific research isdivided into the following three parts:1. Fluorescein derivants (Fluorescein, Fluorescein-DA and Fluorescein-DA-Fe)with three-ring planar structure is used to investigate the interaction with BSAmolecules and the damage effect to the BSA molecules under ultrasonic irradiation,fluorescence and UV spectroscopy were used as the mainly detective method. TheFe(III) ions in Fluorescein-DA-Fe makes it having the additional sonocatalyticdamage effect to BSA molecules. Three-dimensional fluorescence spectra andthree-dimensional fluorescence contour profile spectra were used to investigate thefluorescence quenching effect and the conformational changes of BSA molecules in the presence or absence of Fluorescein derivants. The degree of injury and mode toBSA molecular were examined by a number of factors such as: ultrasonic irradiationtime, Fluorescein derivants concentration and the NaCl concentration). Finally,Oxidation-Extraction Spectrometry was used to detect the types of ROS produced indamage process of BSA molecules.2. This section mainly uses fluorescence spectroscopy, UV spectra, synchronousfluorescence spectroscopy and circular dichroism to examine the interaction of CresolRed derivants (Cresol Red, Cresol Red-DA and Cresol Red-DA-Fe) with BSA. Basedon these, the corresponding spectroscopic techniques were also used to study thesonodynamic and sonocatalytic damage of Cresol Red derivants to BSA moleculeunder ultrasonic irradiation. While some factors ultrasonic irradiation time, CresolRed derivant concentration and ionic strength are also used to study the damage ofBSA molecules. In addition, the synchronization spectrum can be given to researchthe binding sites and the damage sites of Cresol Red derivants to BSA molecule.Finally, the Oxidation-Extraction Spectrometry was used by us to examine the type ofreactive oxygen species (ROS).3. The Thymol Blue derivants (Thymol Blue, Thymol Blue-DA and ThymolBlue-DA-Fe) were used as sonosensitizer after irradiated by ultrasonic irradiation.Firstly, the interaction of Thymol Blue derivants with BSA molecular is examined byfluorescence spectroscopy. The sonodynamic and sonocatalytic damage degree of theBSA molecule taken by Thymol Blue derivant under the ultrasonic irradiation wereexamined by ultraviolet spectroscopy, fluorescence spectroscopy and circulardichroism. Here, the ultrasonic irradiation time and Thymol Blue derivantsconcentration are the main factors to examine the damage changes. The types ofOxidation-Extraction Spectrometry are still used to examinate the types of ROSgenerated in sonodynamic and sonocatalytic damage process.The polycyclic compounds mentioned in this paper, as well as the polycycliccompounds after adding amino acids and metal ions in the treatment of tumors, laid atheoretical foundation for the future research.
