Mercury is a kind of heavy metalpollution, and its organic state-methyl mercury toxicitymore intense. The methylmercury has the very strong neurotoxicity, along the animal foodchain, the skin and breathing three forms into its body, through to the biological organismspoison enrichment, biological amplifier and biological accumulation effect in the animalaccumulation, make the body than the concentration content of survival environment, andcause irreversible damage.In this paper, the Jipagou gold mine is in the Huadian city Jilin province, the gold miningcause serious pollution to the local environment. Paper chose the Rana Chensinensisthat isrepresentative amphibious animal oflocal river-forest wetland system, as the research object.At same time, pater collects gold mining region river water and sediment as environmentsamplings. The paper analysis the methylmercury distribution characteristics and measuresDNA methylation level of the Rana Chensinensis body part.The experiment is divided into two parts: in the first part, the methylmercury contentwere determined and analyzed of Rana Chensinensis different parts of the body, analyzed therelationship among various parts, methylmercury content of Rana Chensinensis, weight,length and the local environment. In the second part, the DNA methylation level weredetermined and analyzed in every part of the body Rana Chensinensis, and analyze the impactof changes in the DNA methylation level factors.The results show that:1. thestudy area gold mining has caused severe local environmentmercury pollution. The river water methylmercury is moderate level of contamination, thesediment methylmercury is high environmental risk;2. The Rana Chensinensismethylmercury of Wu daogou is the highest level in the study area,342.35ng/g, the RanaChensinensis methyl mercury of Er daogou is the least level in the study area,170.82ng/g, Themethylmercury content of the brain is the highest part in the Rana Chensinensis body,64.72ng/g, the methylmercury content of eggs is the least part,19.64ng/g;3. TheRanaChensinensisbody weight, the length and the methylmercury content have significant positivecorrelation, the methylmercury of Rana Chensinensis and the environment methylmercuryhave the positive correlation;4. The DNA methylation level of Rana Chensinensis in thestudy area is lower than the DNA methylation level of Rana Chensinensis in the control area,the female Rana Chensinensis’s DNA methylation is higher than the male RanaChensinensis’s. On the individual-level point of view, the brain’s DNA methylation is thelowest level, and the muscle’s DNA methylation is the highest level;5. The mainfactor thataffect DNA methylation of Rana Chensinensis is the individual methylmercury content.Themercury pollution inhibits the DNA methylation.Analysis from two aspects: Rana Chensinensis methylmercury content and DNAmethylationin the body, the mercury pollution of the Jipigou gold mining district is serious,has been a threat to the survival of Rana Chensinensis.
