| Lonicera edulis contain a variety of amino acids and sugars, organic acids, vitamins, microelements and minerals, as well as a variety of biological functions, however, the poor stability restricted its application. To improve this situation, the paper applied enzymatic acylation to modify its structure, aiming to improve its stability. Then the study of stability. oxidation resistance, toxicity testing of the acylated products were carried out. to provide a reference in detail.(1) Research of enzymatic acylation.Through the single factor and response surface optimization of time, the ratio, temperature and pH to select the hydroxy acid as acyl donor, under the optimal conditions, the preservation rate of the anthocyanin derivatives could be up to93.51%.(2) Research of anthocyanin derivatives’ purification. Through the response surface optimization experiments of static and dynamic adsorption, I concluded the optimum process parameters.adsorption conditions:sample absorbance0.88nm, adsorption speed1.0mL/min sample volume200mL; desorption conditions were as follows:ethanol concentration47.95%, desorption speed,1.44mL/min, ethanol dosage759.63mL; under this condition, the adsorption and desorption rate reached94.69%, and85.72% respectively.(3) Structure analysis.of the anthocyanin derivatives. Through the ultraviolet and infrared spectra scanning, the characteristic peaks generated on a characteristic absorption wavelength compared to the samples without acylating,which proved the success of acylating.(4) Stability of the anthocyanin derivatives. In order to evaluate the stability of sample, influence of time, pH, light, temperature, metal ions, oxidizing agents, reducing agents, sugars and food additives were invested.The results showed:the stability of the anthocyanin derivatives behaved far better than the samples without acylating.(5) Antioxidance of the anthocyanin derivatives. To research the antioxidant activity of anthocyanin derivatives,the following experiments were carried out, including the scavenging effects on DPPHK·ã€OH and O2·ã€The results showed that:anthocyanin derivatives had far stronger antioxidant ability than the sample without acylating and had certain dose effect relationship.(6) Safety toxicology evaluation of the anthocyanin derivatives. Mice acute and30d feeding tests.had been carried out The results showed that:LD50was greater than10.0g/kg.bw in the fommer test, according to the acute toxicity grading standards, nontoxic. No abnormal performance was found in the later test. For the mice in each group, the weight, the organs, hematological parameters had no significant impact,which proved this transformation was safty. |
