In the present work, firstly, the extraction of flavonoids from Taxus by supercritical CO2fluid, then Optimized supereritical fluid extraction Process conditions. UV and UPLC are the determination method of flavonoids:(1) The working curve of quercetin:y=1.7822x-4.2832, R2is0.9989, the concentration of flavonoids between20μg.ml-1to200μg.ml-1;(2) The work curve of amentoflavone:y=1.4317x-49.013, R2is0.9967, the concentration of flavonoids between63μg.ml-1to630μg.ml-1;(3)The work curve of ginkgetin: y=1.1579x-13.09, R2is0.9989, the concentration of flavonoids between22μg.ml-1to110μg.ml-1; The precision, stability and the rate of recovery of each method has good feasibility, it means all the method is an effective, high extraction rate and shorten time extraction method.The effects of extraction time, extraction pressure, extraction temperature, and ethanol concentration were explored. A second order quadratice quation for supereritical fluid extraction of flavonoids were built and combined with the practice of the actual operation, By using the new method discussed above:(1)The optimum extracting condition of quercetin was obtained as follows:46℃,24MPa,2.3h,82%ethanol, the extraction rate of flavonoids is3.728mg.g-1;(2)The optimum extracting condition of amentoflavone was obtained as follows:48℃,25MPa,2.02h,78.5%ethanol, the extraction rate of flavonoids is4.467mg.g-1;(3)The optimum extracting condition of ginkgetin was obtained as follows:48℃,20MPa,2.38h,82%ethanol, the extraction rate of flavonoids is3.469mg.g-1. At last, the model established by Design-Expert can reflect the extraction process and provide advantages for other flavonoids of the taxus with supercritical CO2fluid extraction. |
