| In nature, straw is a kind of most important regeneration resource, b’ut it can tbe fully used at present because of the difficulty in degradating and transforminglignin. It was found that mixed culture two or more microorganisms can get thebetter degradation effect of lignin after long-term experiments and productionpractice. Now the mixed culture of microorganisms has been widely used for thelignocellulose degradation. The biodegradation of straw by fungi is mainly throughthe enzyme by them, those enzymes can be used to measure the degradation abilityof fungi to straw. The GC-MS is efficient to separate and identify the mix compound,and the FTIR spectroscopy can resolve the problem about molecular structure andchemical constitution, so it is helpful to use the two technology at the study oflignocellulose biodegradation.We also researched the biodegradation mechanism of pure culture and mixculture on rice straw through eluent GC-MS analysis and FTIR spectrum analysiswith rice straw biodegraded in different day in this paper. Through syntheticalanalysis rely on these two detection technology, we can know the basic process ofrice straw biodegradation by the mixed culture of two specific fungi is that firstlysurface wax and the saccharides in the system, a small quantity of cellulose andhemicellulose lignin are degraded, some lignin was degraded to be the monomer andthen both of them degraded by fungi, following the benzene rings are melted andthen lignin is degraded. From analysis of the GC-MS,we know that theidentifications of the mix culture include the pure cultures.The more changes of theintensity of lignin associated bands (at1630,1510and1265cm*1, hemicellulose andcellulose associated bands (at1460,1420,1058and830cm*1), and the relativeintensity of C=0,0H and COOH characteristic bands, the degree of cellulose andlignin degradation become better. We can find that the degradative power of the mixculture is stronger than the pure culture’s after contrast the infrared spectrogram ofthem. After detected the enzyme activity of each plate, we find the result canconfirm the basic process of rice straw biodegradation by the mixed culture to acertain degree, the activity of MnP or LiP don’t have the absolute connection withthe degradation ability of co-culture, and their interrelation remains furtherresearch. |
