Pyruvate, the anionic form of a simple a-keto acid, plays a key role inintermediary metabolism, being the end-product of glycolysis and the startingsubstrate for the tricarboxylic acid (TCA) cycle. Pyruvic acid and its ester have beenreceiving increasing attention with regard to applications in the food, flavor, andpharmaceutical industries, because it has several biological functions such as againstoxidant damage, enhance exercise performance and improve body composition,decrease in body fat, inhibits the growth of hepatic tumors, Anti-inflammatory et al.Ethyl pyruvate (EP), a stable aliphatic ester derived from pyruvic acid, has a strongfragrance, so it is improver to be added to food. Calcium pyruvate is derived frompyruvic acid, often as a weight-loss dietary supplement, it dissolves readily in waterand would not be used in oil and fat. Therefore, it is essential to develop a new kind ofpyruvate derivative as the functional fat and oil used in Cold oil, cocoa butter andchocolate coating.Pyruvated structolipids is not only has the characteristic of short-and long-chain acyl triglyceride molecules, but also has more function than traditionalones. Pyruvated structolipids is a kind of new low caloric fat which has weight lossfunction, also a kind of new cocoa butter substitute which has the function ofenhancing exercise performance, so its exploitation has good economic benefit.With the p-toluenesulfonic acid as the catalytic agent, the process conditions ofpyruvated structolipids produced by glycerol monolaurate and glycerol. The productwas analyzed by Thin-Layer Chromatography methods and HPLC/ES-MS. Anindirect measuring method is introduced in this paper, which has been applied to thesensor the conversion of pyruvic acid without standard. We use the way of theintegration of liquid-liquid extraction and acid-base titration technology to analyzeconversion of pyruvic acid. In this method, hydrolysis of the pyruvated structolipids isprevented. This method also required less power, simple to operated, with higher reproducibility and can be applied in the quantification in lab. As the best solvent,ethyl acetate was selected and determined through theoretical analysis. According tothe theory of extraction efficiency, the times of extraction should be controlled at8.The effect of reaction time, concentration of catalyze, vacuum degree, stirringmethod and reaction temperature on conversion of pyruvic acid were investigated,respectively and optimized by response surface methodology. Studies show thatvacuum degree is unrelated to conversion of pyruvic acid and conversion couldincrease when pressure is low enough. The optimized technology of the preparationwas:reaction time3h, concentration of catalyze3.5%, reaction temperature60℃. Andthe maximum conversion of pyruvic acid was64.8%. This method possessed suchadvantages as short reaction time. environmentally benign procedures and easypurification.
