Carotenoids is a general name for a kind of nature produced. It is found abundantly in plants, microalgae, microbial and sea animals. Carotenoids has an effect of antioxidant, anti-cancer, anti-aging and so on. It is well applied in field of medicine, food, chemical and agriculture.The content and composition of carotenoids in sea cucumber oven was studied in this paper. Extraction technology include of organic solvent extraction, ultrasonic wave assisted extraction, saponification extraction, enzyme extraction and fermentation extraction, and the extraction rate was comparised.Firstly, the qualitative analysis was carried out by TLC. The result shows that there is a proportion of astaxanthin and three other contents in sea cucumber oven. The method of spectrophotometry showed the content of total carotenoids in sea cucumber oven is113μg/g. The content of astaxanthin is23μg/g analyzed by HPLC, which accounting for20.3%of the total carotenoids.Secondly, the technology of organic solvent extraction, ultrasonic wave assisted extraction and saponification extraction was optimized. The conditions of organic solvent extraction technology were using ethyl acetate, extract temperature40℃, extract time3h and the ratio of solvent tomaterial1:10. By above extraction technology the carotenoids content was63.2μg/g. The conditions of ultrasonic wave assisted extraction were ultrasonic power70W, ultrasonic time40min and the carotenoids content was70.1μg/g. The conditions of saponification extraction were using0.2M NaOH, addition amount2.4咪g/g, saponated time50min and saponated temperature40℃,the carotenoids content was70.1μg/g.Finally, the condition of fermentation extraction was explored. The microorganism method conditions were using aspergillus848, inoculation amount of bacteria6%, fermentation time3d. the carotenoids content was85.5μg/g.The enzyme reaction extraction condition were using lipase, amounts of enzyme8%, reaction temperature35℃and reaction time35min. the carotenoids content was92.6μg/g. By contrast of these extract methods, the fermentation extraction is the most effective method. The extraction rate of microorganism method is lower than enzyme reaction extraction, followed by ultrasonic wave assisted extraction, saponification extraction. The lowest method is organic solvent extraction. In view of every factor, fermentation extraction is a feasible way to extract carotenoids from sea cucumber oven in industrial scale.
