Grape seed is byproducts of grape, accounting for about5%of the weight ofgrapes. Grape seed fat content of about14%-17%, linoleic acid content of up to68%-76%. Proanthocyanidins has strong antioxidant, antioxidant, free radical scavengingcapacity of up to50times more vitamin E, and20times as vitamin C.OPC has astrong inhibitory effect as well as a variety of enzymes such as pancreatic lipase, theenzyme tyrosine,α-amylase.In this paper, the use of supercritical CO2fluid extraction of grape seed oil grapeseed powder as the raw material, the use of organic solvent extraction and separation ofmacroporous resin grape seed proanthocyanidins, explore grape seed proanthocyanidinson the inhibition of pancreatic lipase activity, determine its inhibition type andinhibition constants.Grape seed powder skimmed proanthocyandins preliminary extraction. Byorthogonal experiment to explore different factors such as solvents, solid-liquid ratio,extraction time, extraction temperature, solvent concentration on proanthocyanidinsextraction efficiency. Orthogonal test results to determine the best proanthocyanidinsextraction process plan, namely solid-liquid ratio1:10,70%ethanol,2hours, theextraction temperature70℃, extracted three times, In the process conditions, the extractprocyanidins content is4.94%. Through this process has the advantage of highextraction rate of proanthocyanidins, safe, is easy to operate.Process Purification grape seed proanthocyanidins, using macroporous resinspecificity of proanthocyanidins adsorption and desorption were purifiedproanthocyanidins. Static test screening from8different resins the best resin HPD100resin, resin adsorption capacity proved HPD100grape seed proanthocyanidins,desorption rate, the spray dry high purity, while the large mechanical strength of theresin, acid conducive to grape seed proanthocyanidins industrialized separation. Byadsorption experiments to determine the optimal concentration of procyanidin sample8.0mg/mL, the sample pH is3.0, the flow of the sample was3.0BV/h. Volume ofwashing water washing4.0BV, with60%ethanol desorption2.0BV/h flow rate ofelution proanthocyanidins dried purity obtained by the process for more than95%.Grape seed proanthocyanidins inhibit pancreatic lipase activity by changing thedifferent experimental conditions the enzyme reaction system, investigate the effect of grape seed proanthocyanidins on pancreatic lipase activity. The results showed thatgrape seed proanthocyanidins under certain conditions inhibit the activity of pancreaticlipase, pancreatic lipase proanthocyanidins inhibition constant Ki=0.1032mg/mL,change to a different inhibitor concentration and substrate concentration plottedexperimentally determined by Dixon and Weaver-line diagram to prove thatproanthocyanidins on the inhibition of pancreatic lipase activity are noncompetitiveinhibition.
