Erwinia carotovora subsp. atroseptica strain SCRI1043(Ecaa1043) only caused bacterial blight disease in potato and brought about great economic losses. In the current more and more research aimed to explore the pathogenic mechanisms and tried to fight the root rot. In our analysis, we constructed a protein-protein interaction (PPI) network of Eca1043with two computational approaches (interologs method and domain combination method), and verified by K-nearest neighbors (KNN) method.The predicted PPI network of Eca1043contained31,692interactions among2,370proteins. Topology analysis confirmed the reliability of the network after constructing the network. In addition, we made further analysis of the basic properties of the network as well as some important subset of the network, such as secretion system network、flagella synthesis and chemokine system network、signal transduction network, and analyzed biological functions of Eca1043PPI network. Detailed analysis presented that type II secretion system protein secreted some important enzymes in Eca1043, particularly protease and cleavage enzyme, directly affected its pathogenicity. In addition to the secretion system, we also have found that FliM and FliN were important protein of flagellar synthesis switch, especially FliM, plays an important role for the other flagellar synthesis. Chemokine, especially CheA, associated with many enzymes and proteins, while regulated flagellin with other chemokines. Furthermore, we found that a lot of two-component signal transduction systems in Eca1043, especially HrpY-HrpX system, had a great relationship with its pathogenicity. The predicted PPI network provides valuable clues to explore the pathogenicity and metabolic regulation of Eca1043.
