Greenshell character has always been selected as an important economic indicator. Currently, select green shell character by traditional method.Although the molecular breeding technology continuous development for the pathogenies green shell properties of candidate genes have been reported in recent years, the laboratory research results can be applied to actual production has not yet been reported.This test will be on correlation between SLCO1B3gene and green shell, aims to provide candidate genes of green shell character for jinyun duck.In testcross experiments, according to the pedigree information, select40high performance green shell jinyun duck, as male parent, using a scale of1:20white shell egg jinyun pairing, compose40families (F0), make textcross spring as F1generations. Shell color is controlled by a pair of major gene, green shell color as the dominant character, white shell is recessive.According to F1generation eggshell color and two color shell proportion identify parent drake gene type; After testcross experiment, abandon of a offspring too little family, collected testcross483offspring duck;39drake under test, only6male duck green shell genotype homozygous.In this experiment, make jinyun duck textcross offspring as the research object. Random selecting four produce Green shell egg and four produce white shell egg duck and picking up oviduct tissue. According to the NCBI find chicken SLCO1B3gene conserved region, across exon region design primer, using SYBR Green I real-time quantitative PCR (qPCR) technology, adopt to the method of relative quantitative, with β-actin for internal gene, detection SLCO1B3gene expression in Green shell and White shell individual oviduct tissue,and using2-△△Ct method analysis results; the result show that SLC01B3gene in white shell eggs oviduct tissue position expression quantity is higher than that in the production of green shell eggs, and significant difference (P<0.05), the author thinks that the gene can be used as one of green shell candidate genes.Because of the regulatory regions generally in gene upstream position.In order to reveal the reason of SLCO1B3gene expression difference, we should get SLCO1B3gene5’UTR region sequence information. It is very important to find the green shell gene molecular markers by comparing the different characteristics of individual5’-UTR region. Sequencing results showed the5’-Race technology has a length of320bp gene.NCBI found this gene with chicken LOC422031and LOC422032gene has a similar rate of83%,, speculate that the duck and chicken SLCO1B3gene mutation in the regulatory regions.In summary,40Jinyun duck contain green shell homozygous only6individuals; SLCO1B3gene have different expression in production of different shell color individual Jinyun duck oviduct.By NCBI comparison, found the gene with chicken SLCO1B3gene have similarity. Further studies of the gene, may help to explore Jinyun duck green shell character molecular markers.
