Objective:To investigate the effects of overexpression and knockdown of the regenerating gene (REG) IV on proliferation, migration and invasion of colorectal cancer cells.Methods:REG IV mRNA was quantified by real-time PCR and the expression of REG IV protein was analyzed using western blotting. The cell proliferation was detected with 3-(4.5-dimethylthiazol-2-yl)-2.5-diphenyltertrazolium bromide (MTT) and the expression of proliferating cell nuclear antigen (PCNA) by immunohistochemistry assay. The experiments of migration and invasion were conducted to explore the potential role of REG IV on the invasiveness of HCT-8 cell.Results:Our results showed that the expression of REG IV mRNA and proliferation of HCT-8 cells were increased after transfecting PCDNA 3.1-REG IV (P<0.05), while decreased by blocking of REG IV gene using the gene specific siRNA (P<0.05). Both migration and invasion capacities were found to be profoundly repressed in REG IV RNAi-2 treated cells. And the number of migrated cells was significantly increased after transfection of PCDNA 3.1-REG IV (P<0.05).Conclusion:It indicates that Reg IV is an important regulator of cell expression, proliferation, migration and invasion. Hence, it is a potential target for adjunctive treatments for human colorectal cancer.
