Objective: To observe the effect of thyroid hormone in human umbilical vein endothelialcell proliferation, migration and cell ultrastructure.Method:1.To determine the possible effect of TH on induced human umbilical veinendothelial cell proliferation, we examined the viability of endothelial cells by MTT assay.2. Themigration ability of human umbilical vein endothelial cells was measured with scratch assay, andTranswell chamber.3. We use atomic force microscope (AFM) to observe the changes ofultrastructure on human umbilical vein endothelial cell surface treated with different levels ofTH.Results:1.The data clearly showed that TH can induce HUVECs proliferation,cellproliferation and TH concentrations showed dose effect relationship.2.TH promotes migrationof HUVECs, HUVECs migrated longer distance which treat by high concentration of TH.Transwell chamber assay showed that the cells passing the chamber of the group that treated inhigh concentration of TH was significantly more than in the control cells and in lowconcentration of TH treated cells (P<0.05).3.AFM was used to examine the changes ofultrastructure on HUEVCs surface treated with different concentrations TH.After the treatmentwith TH,there were many pseudopodium and the granules extended the edges of cellobviously.There were significant difference between the group of treatment with different THlevels.Conclusion: Thyroid hormone can promote HUVECs proliferation, migration,also changethe ultrastructure of the HUVECs surface.
