Study On Anti-tumor Metastasis And The Influence Of The Nude Mouse Transplantation Tumor Growth Effect Of Curcumin Derivative-C086

Objective To provide the basis for the anti-tumor effect of curcumin derivative–C086in vivo and in vitro.To explore the anti-tumor metastasis effect and the mechanism of itsanti-tumor metastasis effect.Methods （1） To investigate the effects of C086on proliferation of gastric cancer cellsSGC-7901in vitro by MTT assay.（2）Using clone formatin assay to detect the activity ofproliferation inhibition of C086on SGC-7901.（3） Scratch test and Transwell assay wereperformerd to detect the effect of C086on migration ability and invasion ability ofSGC-7901.（4） Western blot assay was used to test the protein level of MMP-2andMMP-9on the C086worked SGC-7901cell.（5） Established the Kunming mouse modelof H₂₂ experimental pulmonary metastatic model in vivo,and evaluate the anti-tumoractivity of C086in vivo.（6） To evaluate the anti-tumor activity of C086in nudemice,SGC-7901human gastric cancer was established in nude mice.（7） Western blottingwas used to analyze the protein level of Erk1/2, C-Mycand Bcl-2of the xenograft tumorof nude mice.（8） Using the flow cytometry instrument to detect effect on human gastriccancer cell apoptosis with C086worked SGC-7901cells.Results The proliferation inhibition of C086on SGC-7901had a dose-and time-dependent relationship. The IC₅₀ values to SGC-7901for48h and72h were5.29μg/mland4.17μg/ml.（2）With the increasing concentration of C086, Colony forming inhibitionrate increased gradually and had a does-dependent in some degree. The IC₅₀ values0.55μg/ml.（3）The migration ability and invasion ability of SGC-7901become lower withthe control group by the series of concentration of C086worked cells.The Invadeinhibition rate of2μg/m、4μg/m、8μg/ml C086reached50.65%、72.73%and89.61%（P<0.05）.（4） The protein level of MMP-2and MMP-9on the C086worked SGC-7901cell all become lower.（5）With C08650mg/kg、C086100mg/kg、C086200mg/kg takeedorally, the Kunming mouse model of H₂₂ experimental pulmonary metastatic modelnodules inhibition rate reached28.0%、41.5%and59.7%. Through the lung tissue sliceobservation HE staining,we found that C086protect the lung tissue well, oppose theinvading of tumor cells, controling the tumor cells and metastasis to the lungs.（6） Thetumor inhibitory rates of C086micro emulsion100mg/kg and150mg/kg on the nude mice SGC-7901human gastric cancer after oral daily dosing were52.3%and57.2%.（7）Western blotting result showed that different tumor piece of tissue in nude mice of theprotein level of Erk1/2, C-Mycand Bcl-2Content reduced,and also showed adoes-dependent in some degree.（8） C086can induce SGC-7901middle-late and lateapoptosis.Conclusion The experimental results make it clear that C086had Anti-tumor activity ofinvasion and metastasis in vivo and in vitro.
