| ObjectiveTo observe the expression level of MMP-13, and discuss the possible mechanismof OPG protecting chondrosarcoma cell.Methods1. Culture and subculture the chondrosarcoma cell (SW1353).2. The experiment contain8groups, compare group: the culturing of chondrosarcoma cell;OPG group1-7: stimulating the chondrosarcoma cell by OPG(6.25,12.5,25,50,100,200,400ng/ml).3.Use Western blot to detect the expression of matrixmetalloproteinase-13(MMP-13) chondrosarcoma cell after cultured24h in every group. Divide target protein gray value to the internal reference GAPDF gradation value in order to correct errors, the results represented the relative content of the target protein in a sample. The statistical results are expressed as±SD. Using SPSS19.0to complete the statistical workss. Comparison amonggroups using one-way ANOVA analysis, heterogeneity of variance using rank sum test, P<0.05was considered having significant difference.ResultsApart from the OPG levels of50ng/ml group, OPG between groups of MMPS-13expression amount compared with the blank control group, P <0.05, statisticallysignificant. The inhibition of MMP-13is also enhanced with the OPG concentrationmultiplication. OPG compared between each group:50ng/ml compared other groups,P>0.05, no statistical difference; The rest of the group, P <0.05, statisticallysignificant. Conclusions1. OPG can inhibit the expression of MMP-13in chondrosarcoma cell.2. In different concentration OPG (6.25,12.5,25,50,100,200,400ng/ml),the inhibition of MMP-13is also enhanced with the OPG concentrationmultiplication. |
