| Objective The study was to investigate the effects of rosmarinic acid(RA)on rat glomerular mesangial cells apoptosis induced by Lps,involvedunderling molecular mechanism of glomerular disease.Methods After two months of breding male rats, obtain high purity ofglomerular mesangial cell. Mesangial cells were identifided by invertedmicroscope and immunohistochemistry.Rat mesangial cells were cultured inRPMI-1640with different does of RA(5ã€25ã€50μg/ml) for48h.Cellproliferation by LPS was determined by CCK-8. QRT-PCR indirectimmunofluorescence and ElISA wasUsed to tested the effect of RA on Bcl-2ã€Bax mRNA and proteinExpression in glomerular mesangial cells.Result1. After24h of incubation,the glomerular mesangial cells treatedwith lps displayed features consistent with apoptosis.Such as nuclearcondensation and fragmention,Apotosis rate was significantly higher thanthat of control group(p<0.01)2.RA can obviously induce apoptosis of GMCin a dose-dependent manner to some extent.The expressions of Bcl-2weredecreased and Bax were increased after incubating with RA,the ratio of BcL-2and of Bax was significatntly decreased.Conclution1.RA induces apoptosis of rat mesangial cells in does-dependentmanner.Its mechanism may be that apoptosis of MC result from altered ratioof Bcl-2and Bax level by apoptosis down-regulation or up-regulation ofcontrol gene. |
