Establishment Of The Rat Model Of Tuberculous Pleurisy And Pathological Research Of Membrana Pleuralis

[Objective] The present study is aimed at establishing tuberculous pleurisy model in rat and observing the membrana pleuralis pleurisy pathological change of berculous pleurisy in different time with eye, light microscope and transmission electron microscope.[Methods] The study select30Wistar-rats as research target,trabant choose26rats as eperiment group,4rats as control group. The30rats simultaneous inoculation Bacillus almette-Guerin suspension0.1ml(0.06mg), we will do pleuracentesis after five weeks. We will influx standard Mycobacterium hominis H37RV mycobacterium tuberculosis suspension1ml into experiment rats, and establish tuberculous pleurisy animal model. After establishing animal model, we will kill the rats in batch at2d、4d、6d、8d、10d、12d、14d、16d、18d、22d、30d, two rats every time. We influx I sotonic Na chloride2ml into normal rats,and put it to death after2d,4d, two rats every time. After executing the rat,we expose their thoracic cavity, review the pathological change of membrana pleuralis.[Results] The amount of pleural effusion increase gradually in earlier period, the amount add to peak at6-8d, then taper, at16-22d most pleural fluid(PF) disappear. In earlier period pleura(PLE) engorge and dropsy, fibrin(FIB) exud, lymphocyte infiltrate into the bottom of pleura(PLE), we can see epithelioid cell among the group of leukomonocyte, On the pleura there are Focal necrosis and mesothelial Cellular proliferation; We can see coagulation necrosis、granuloma scobination and mesothelial cell damaged by degrees. In later stage we can see pachynsis pleurae、accretio、tubercler and anabrosis focus on the pleura. Under the light microscope there are epithelioid cell clump increasing、tubercler、fibroplasias and caseous necrosis. Under electron microscope mesothelial cell membrane and cell-substance segmental defect, it is show up that fibroblast and fibrocyte、desmoplasia under the mesothelial cell, there are most collagen fibers that arrange fasciculation irregular.[Conclusion]1.The rat model of tuberculous pleurisy was established by infusing standard Mycobacterium hominis H37RV into rat after inoculated BCG five weeks.The observation of membrana pleuralis pathology is consistent with the distinction of tuberculous pleurisy, the acid-fast bacilli was discovered in the necrotic tissue by acid fast stain.2. The pathological change of tuberculous pleurisy:It is mainly exudation in early period, there are phlegmonosis cells infiltrating, cell organelle are destroyed in different degree, then mesothelial cells are destroyed, it is composed tubercler that collagen fibers accrementited, macrophage transformated into epithelioid cell、Langhans giant corpuscle, it is appeared that caseous necrosis, the three patho-change:can exist at the same time.
