| Tea flower, as one of reproductive organ from Camellia sinensis, contains many kindsof bioactivity materials, such as protein, tea polysaccharide, amino acids, vitamins, etc. Ithas been reported to have effects on detoxifcation, lipid-lowering, anti-cancer, and beautymaintenance, etc. This research studied on the extraction and purification of tea polyphenoland tea saponin from tea flowers using ultrasonic wave method, and researched the relatedbacteriostasis. Then the bioactivity materials can be developed and utilized from teaflowers more efficiently, which provide theoretical foundation for the comprehensiveustilize of tea flowers.Theasaponins, which is widely distributed in the root, leave, seed and flower of teaplants, a mixture of olean-type triterpenoids saponin that roots in theaceae camellia plants.Theasaponins, as a good natural surfactant, can remove freck, diminish inflammation,relieve a cough, ease pain, resist penetration, disinfect, kill insect, promote plant growth,etc.Tea polyphenols (TP), also called “Tea tanninsâ€, is a kind of mixture of polyphenols. Itis not only a natural antioxidant, but also an ideal medicine, which have so many healthfunctions and pharmacological actions as resisting oxidation, antivirus, bacteriostasis,anti-radiation, cancer-fighting and so on.Ultrasonic extraction was adopted to extract theasaponins and polyphenols from teaflowers, Firstly the parameters such as the ethanol concentration, extraction temperature,ratio of solid to solution, ultrasonic power and extraction time, which effected on theintegrated extraction were studied by the single factor experiments. Then this paper utilizedthe orthogonal experiment to optimize the technological parameters of the comprehensiveextraction, which included ethanol concentration (A), extraction temperature(B), ratio ofsolid to solution(C), ultrasonic power(D) and extraction time(E),and the factors wereanalyzed by range and variance analysis. The results showed that the significant factors ofinfluencing the yield of tea saponin as follow: A, D, B, E and C, and the significant factorsof influencing the yield of tea polyphenol as follow: A, B, D, E and C.By thecomprehensive consideration, the optimum extraction conditions as follows:70%ethanolconcentration,350W ultrasonic power,60℃extraction temperature,1:30g/mL ratio ofsolid to solution and10min extraction time. Under the condition, the yields of tea saponinand tea polyphenol were up to23.69%and28.38%,and the results were reliable.Comparing the resins HPD100, HPD722, AB-8, D101and DM130by staticadsorption, the resin DM130was chose. After finishing chromatography column, sample was eluted with different concentrations of ethanol, then two kinds of condensed and driedwere gotten. Through spectrophotometric determination, the purity of tea saponin and teapolyphenol was90.9%and87.3%, respectively.Through the study of tea saponin and tea polyphenol antibiotic effect toStaphylocooccus aureus, Bacillus subtilis, Micrococcus tetragenus and Yeast, it showedthat tea saponin and tea polyphenol had antibiotic effect to the four germs, especially teasaponin had strong bacteriostasis to Yeast. |
