Objective: Exploring the expression change of genes and their relevant miRNAsin apoptotic leukemia cells induced by Demethylcantharidin (DMC).Methods: After DMC inducing K562cell apoptosis, microarrays assessment wasperformed to detect the expression changes of genes and miRNAs. RT-PCR, real-timePCR and enzyme-linked immunosorbent assay (ELISA)were performed to analyze thecorrelation between genes and their relevant miRNAs.Results: Microarrays results showed that more than2300genes transcripts and290miRNAs were expressed differently after DMC treatment. Tumor suppressor genes(RB1and P53)were up-expressed and oncogenes (BCL2, E2F1and E2F3) weredown-expressed. miR-16, miR-34a, miR-34b, miR-34c, miR-17and miR-125wereincreased more than1.5times, while miR-20, miR-106and miR-150weredown-expressed more than1.5times. RT-PCR, real-time PCR, and ELISA analysis alsosupported the changes.Conclusions: Our study showed that there were negative correlation between theexpression of miR-16/34a-c and BCL2, miR-17-5p and E2F1, miR-125and E2F3,miR-106and RB1, miR-150and P53. These miRNAs may regulate the expressions ofthe above tumor suppressor genes or oncogenes.
