[Objective]To observe the expression of Pim-3in lung adenocarcinoma A549cell line and in different pathological types and different TCM syndromes of lung cancer tissues, and discuss it’s clinical implication.[Methos]1. Collection of specimens:27paraffin-embedded postoperation specimens keeping by the pathology department of Shiyan Taihe Hospital were collected by us. Diagnose them through interrelated diagnostic criterias presented by Chinese Medicine Association. Divided them into different pathological types, including6cases of non-cancer bronchial and pulmonary tissues,7cases of lung adenocarcinoma,7cases of lung squamous carcinoma,7cases of small cell lung cancer. Each specimenvwas cutted intotwo slices. And we also collected lung adenocarcinoma A549cell line cultured in vitro for the experiment.2. Test method:Each group of specimens were examed by Immunohistochenmistry staining after dewaxed, thus test expression of the Pim-3protein in cells. Observing cells under light microscope, We judged that it’s positive if endochylema in cell was yellow-dyed even and dense, and else, it’s negative if endochylema in cell was not obviously yellow-dyed.5high power visual fields (X400) were random selected in every section, counted positive-presented cells and total cell amount under visual field, figure up every specimen’s positive-expression rate, thus compute every groups’ positive-expression rate. Compare diffrences between every two groups with x2-test, compute the p-value with SPSS11.0software. Analyse collected lung adenocarcinoma A549cell line cultured in vitro, different pathological types of lung cancer tissues, and non-cancer bronchial and pulmonary tissues. Examining their expression of Pim-3by immunohistochenmistry, analysing expression differences among A549cell,non-cancer bronchial and pulmonary tissues, and differente types of lung cancer tissues.[Results]1. After immunohistochenmistry staining, it was seen yellow-dyed even and dense in lung adenocarcinoma A549cells, the positive expression rate was100%.2. After the12slices of non-cancer bronchial and pulmonary tissues were dealed with immunohistochenmistry staining, no yellow-dyed was seen in alveolus pulmonis epithelial cells, smooth muscle cells, vascular endothelial cells and bronchial epithelial cells’ cytoplasm. Positive expression rate of Pim-3protein was0.3.3. In7cases of lung adenocarcinoma,7cases of lung squamous carcinoma,7cases of small cell lung cancer, with a total mount of42sections, We’ve seen yellow-dyed even and dense in cytoplasm in varios typs of cacer tissues. Positive expression rates of Pim-3were all100%. And we compared every two groups with x2-test, the p-values were all>0.05, no obvious differences were found. However, no yellow-dyed was found in the surrounding-cancer tissue cells.[Conclusion]After dealed with immunohistochenmistry staining, the positive expression rates of Pim-3protein were all100%in A549cells cultured in vitro and in various types of postoperation lung cancer tissue specimens. However, it’s0in surrounding-cancer tissues and non-cancer bronchial and pulmonary tissues. The specificity high expression of Pim-3protein in lung cancer tissues can separate lung cancer from non-cancer lung diseases. These indicate that Pim-3protein probable be a specificity biotic marker in the process of lung cancer’s occurring and progressing. The detection of Pim-3may be used at clinical diagnose of lung cancer.At the same time, Pim-3’s positive expression rates were all100%in different pathological types of lung cancer tissues. No obvious difference was found among them. Therefore, although Pim-3probable could be used at clinical diagnose of lung cancer, it can’t distinguish different histological type of lung cancer.
