Objective:To explore the effects and mechanism of rhubarb on intestinal microcirculationin septic rats,by observing the change of intestinal blood flow and oxygenation in intestinaltissue.Methods: Fifty-six healthy male SD rats with weighing220-250g were randomlydivided into control group,sham group,sepsis group,and rhubarb group.we emplyed thececal ligation and puncture (CLP) model.The rhubarb group were gavaged with rhubarb(50mg/kg) twice a day for two days before CLP septic modeling.And the other groupswere gavaged with normal saline instead. We detected the blood flow of intestinal mucosaand wall twenty four hours after modeling with moorFLPI laser speckle imaging system.Assaying the concentration of lactate and pyruvic acid in homogenated supernate ofintestinal tissue with ELISA and calculating the ratio of L/P (lactate/pyruvic acid).Todisplay the capillary in intestinal mucosa with perfusing gelatin and ink and HE stain, andthen compute microvascular area (MVA). Observing the CD31expression withimmunohistochemistry and counting microvascular density (MVD). Results:(1)Compared with sepsis group(2020.3±353.4), the blood flow of intestinal wall in controlgroup (2799.4±370.6,p<0.01), sham group (2514.2±484.9,p<0.01) and rhubarb group(2653.0±284.0,p<0.01) are much higher (unit:pu).(2) also, Compared with sepsisgroup(1662.6±462.0), the blood flow of intestinal mucosa in control group(2548.2±336.5,p<0.05), sham group(2514.0±565.1,p<0.05) and rhubarb group(2383.2±834.9,p<0.05) aremuch higher (unit:pu).(3) The L/P ratio in intestinal tissue are taller in CLP sepsisgroup(0.64±0.03) than control group(0.52±0.05,p<0.01), sham group(0.43±0.04,p<0.01),and rhubarb group(0.50±0.02,p<0.01).(4) The area of intestinal mucosa capillary in sepsisgroup (3.68±1.13) are less than control group (6.26±0.69,p<0.01), sham group(6.04±0.37,p<0.01) and rhubarb group(6.73±1.52,p<0.01).(5) Compared with the counting ofcapillary on intestinal mucosa in sepsis rats(41.08±5.90), the control group(59.42±5.43,p<0.01), the sham group(55.08±5.80,p<0.01) and the rhubarb group(56.87±6.36,p<0.01)are more.Conclusion:The rhubarb can expand the intestinal mucosa capillaries, reducethrombosis, protect intestinal mucosa capillary endothelial cells, andincrease the number of intestinal functional capillary. Thus, it may improve the supply ofintestinal blood and oxygen with sepsis.
