The Prognosis Of Primary Cutaneous Malignant Melanoma In Clinical Analysis

Objective: To study the relationship between methods of treatment of primarycutaneous malignant melanoma,ulceration or not, serum LDH level, the result of theimmunohistochemical of S-100、HMB-45and Melan-A of cancer tissues, gender, ageand2-year survival of patients.To explore its significance in clinical and whether canbe viewed as aided diagnosis and prognostic factors of primary cutaneous malignantmelanoma.Methods:1.Selected the clinical data of primary cutaneous malignant melanomaof144patients in the Liaoning Provincial Tumor Hospital during January2006-January2009.2.Mouse anti-human S-100monoclonal antibody, mouse anti-human HMB-45monoclonal antibody, mouse anti-human Melan-A monoclonal antibody and secondaryantibodies were purchased from Beijing Zhongshan Golden Bridge BiotechnologyCompany.3.All of the specimens were made in the common way,and the expression ofS-100protein, HMB-45protein and Melan-A protein was detected withimmunohistochemical methods.①After with4%formaldehyde fixed,all of thespecimens were embedded and cut in the common way, its thickness is5μm.②Dewaxing in the oven with63degrees overnight,all of the tissue biopsies werehydration.③According to the special requirements of primary antibody,all of the tissuebiopsies were dealed with.④All of the tissue biopsies were incubated in3%hydrogenperoxide solution for10minutes,in order to block endogenous peroxidase enzyme.⑤All of the tissue biopsies were flushed with PBS in every2minutes3times.⑥After dropping primary antibody,all of the tissue biopsies were incubated with37degreesover1or2hour.Then they were flushed with PBS in every2minutes3times.⑦Afterdropping secondary antibody,all of the tissue biopsies were incubated with37degreesover20minute.Then they were flushed with PBS in every2minutes3times.⑧Letall of the tissue biopsies to color in DAB solution.⑨All of the tissue biopsies were inthe Water washed, stained, dehydrated, transparent and fengpian.Results:The methods of treatment of primary cutaneous malignant melanoma,ulceration or not, S-100, Melan-A and age exists statistically significant difference inthe control group(P <0.05), while gender, serum LDH levels and HMB-45do notexists statistically significant difference in the control group(P>0.05).Conclusion:The cutaneous malignant melanoma is a high proportion of malignantmelanoma, this study shows:①This is correlation between the methods of treatmentof primary cutaneous malignant melanoma, ulceration or not, S-100, Melan-A, ageand the survival of patients.②After surgery， the survival of patients usedchemoimmunotherapy treatment four times is longer than patients only useddacarbazine.③S-100, HMB-45or Melan-A should be regarded one of waysof aided diagnosis of the cutaneous malignant melanoma and one of indicator ofprognostic of the cutaneous malignant melanoma.
