| Objective: The present study was designed to investigate the effect of β-elemeneon proliferation of hepatocellular carcinoma cell line H22and expression of histone H1.Methods:1ã€The expression of histone H1in H22cells was detected by Western Blot,RT-PCR and immunofluorescence.2ã€The expression of histone H1in H22cells which were treated by β-elemene wasdetected by Western Blot, RT-PCR and immunofluorescence.3〠The expression of histone H1in H22Xenograft Transplantation Model whichwere treated by β-elemene was detected by Western Blot, RT-PCR andimmunohistochemistry.Results:Firstly, we have demonstrated that histone H1was expressed in murinehepatocellular carcinoma cells H22. Next, H22cells were cultured with β-elemene fordifferent time in vitro and treating with different dose-dependent β-elemene in anexperimental H22hepatocellular carcinoma (HCC) xenograft transplantation modelwere to confirm if β-elemene could inhibit the growth of H22tumor cells. In addition,measurements of histone H1expression both in vitro and vivo enabled us to clearlyknow that β-elemene affects histone H1’s expression only in protein level, not involvedin gene level. Finally, our results clearly show that the effect of β-elemene on inhibitingthe growth of H22tumor cells is time-dependent and dose-dependent.Conclusion:β-elemene inhibits the growth of H22cells through enhancing theexpression of histone H1only in protein level. |
