Objective:To investigate the mechanism of glutamate signaling pathway in the filopodias ofepidermal cell and the melanosome transport.Methods:1. After culturing the human epidermal melanocyte and keratinocyterespectively,we use scanning electron microscopy to observe the various changes of themelanocytes and keratinocyte with NMDAR (glutamate receptor) noncompetitiveantagonist MK-801, agonist N-Methyl-D-aspartic acid (NMDA).2. Melanocyte and keratinocyte were co-cultured,then the morphology changeswere imaged by scanning microscopy to observe changes in melanocytes andkeratinocytes when they were treat with MK801、NMDA.3. Melanosome transport were imaged by confocal microscope after the cell treatedwith MK801、NMDA.Results:1. A scanning electron microscope found the MK-801、NMDA will enablemelanocyte morphology change. Melanocytes in the MK801,the end of the celldendrites was significantly narrower and longer,the number of surface filopodias wasless and shorter.In NMDA,the end of the dendrites was significantly wider andshorter, the number of cell surface filopodias significantly higher and longer.2. In co-cultured, we found there are the contact between melanocytes andkeratinocytes in the system, and filopodias of the area close to the direction ofkeratinocytes on melanocytes than in the opposite direction. After the cells treated withNMDA,the filopodias increased but in MK801the filopodias are less.3. Confocal microscopy of fluorescently labeled melanosomes、 keratin, foundthere are melanosomes in the keratinocytes, the number increased by NMDA but less in MK801.Conclusion:1Glutamate signaling pathway can affect morphology and the number of thefilopodias in melanocytes.2Glutamate signaling pathway can affect the transportation of melanosome.3.Glutamate signaling pathway can affect the melanosome transport via thefilopodias.
