Objective To observe the protective effect of triptolide on the expression of nephrin and podocin protein and mRNA in rats with IgA nephropathy(IgAN), and compare the efficacy of different doses of triptolide.To explore the mechanism and curative dose of triptolide protecting podocyte in rats with IgA nephropathy.Methods60SD male rats were randomly divided into normal group, model group, benazepril group, triptolide low dose group, triptolide medium dose group.triptolide high dose group, n=10. In addition to the normal group after the start of the experiment, the remaining rats using bovine serum albumin (BSA)+carbon tetrachloride (CC14)+lipopolysaccharide (LPS) to establish the IgAN rat model, in the eleventh week triptolide and benazepril were given as intervention,the time last for4weeks. Rats were killed at14weekend, specimens were taken from kidney tissue of rats in each group and glomerular mesangial IgA deposition was observed by immunofluorescence staining. In addition, the expression of nephrin mRNA and podocin mRNA were detected by Real-time PCR and the expression of proteins nephrin and podocin were detected by ELISA.Results (1)At first, Model group in Immunofluorescence mirror could be seen IgA immune complex diffuse deposition (see chart1), model group by light microscopy could be seen mesangium severe hyperplasia (see chart2).(2) The expression of nephrin mRNA and podocin mRNA in modle group all became significantly weaker compared with that in normal group (P<0.01);their expression in TP each dose group and benazepril group were stronger than that in modle group (P<0.05) and the difference in the expression of nephrin mRNA and podocin mRNA in the three TP groups and benazepril was not statistically significant (P>0.05);there was no obvious difference among three TP groups (P>0.05).(3) The expression of protein nephrin and podocin in modle group all became significantly weaker compared with that in normal group(P<0.01); their expression in TP each dose group and benazepril group were stronger than that in modle group (P<0.05); their expression in TP medium dose group and TP high dose group were stronger than that in benazepril group (P<0.05);the difference in the expression of protein nephrin and podocin in the TP low dose group and benazepril group was not statistically significant (P>0.05), their expression in TP medium dose group and TP high dose group were better than low dose group, and there was no obvious difference between the middle and high dose group (P>0.05).(4)Statistics showed that there was negative correlation between24-hour urine protein detected in the anterior experiment and the expression of nephrin and podocin protein and mRNA(P<0.01).Conclusion TP each dose group can raise the expression of nephrin protein and mRNA in rats,they also can raise the expression of podocin protein and mRNA. The effect of middle and high dose groups was more obvious. There was no obvious difference between three TP groups and benazepril group in the comparison of nephrin and podocin mRNA expression, however, TP medium and high dose groups were obviously different from benazepril group in the comparison of nephrin and podocin protein expresion. It hints that TP can raise the expression of nephrin and podocin,reduce the injury to podocyte and recover the damage of glomerular filtration membrane barrier,and the regulating action of TP on nephrin and podocin may be better than benazepril.
