Objective Discuss FCRL gene polymorphism and genetic susceptibility of ankylosing spondylitis with the correlation between the severity of disease, at the same time study FCRL interaction of genetic and environmental factors in the disease. For the treatment of ankylosing spondylitis and causes exploration to provide a reference basis.Methods Choose299cases in first affiliated hospital of anhui medical university from2009to2011in Department of Rheumatology for the AS patients group. According to age and gender matched300cases in the first affiliated hospital of anhui medical university medical center for a control. The AS patients in selected collection of epidemiological data, including the general demographic characteristics (gender, age, nationality, occupation, etc.), BASFI index, BASDAI disease activity index, VAS pain index and environmental exposure (smoking, drinking, eating preferences, etc.). Acquisition of ankylosing spondylitis patients and controls the peripheral venous blood of5ml, with EDTA anticoagulant and separation of plasma, salting out method was used to extract DNA of the patient, using the SNaPshot genotyping technology classification identification of the genes. Epidata3.1were used for double entry and error detection, the results of statistical analysis were using SPSS13.0software, inspection level take alpha=0.05. The study of gene environment interactions were used case-only study.Results The genotype analysis demonstrated that one of the FCRL4tag SNPs rs2777963TT genotype may be a risk factor of AS (x2=7.643,P=0.019). The haplotype analysis indicated that there were no significant differences between AS cases and controls. Patients with AS who had rs14335AA genotype had a significantly declined VAS patient’s global assessment scores compared to those with the GG genotype (31.21±26.25VS40.54±25.40,P=0.035) and GA genotype (38.29±24.94VS40.54±25.40,P=0.044), and in locus rs10489674, TT genotype had a significantly increased BASDAI scores compared to those with the CC genotype (4.73±2.43VS3.15±1.61,P=0.003) and CT genotype (4.73±2.43VS2.97±1.71, P=0.001). A significant interaction of G x E between FCRL4gene and drinking was found (OR=2.610,95%CI=1.303-5.228,P=0.006).Conclusions The FCRL4polymorphisms may play an important role in the susceptibility and severity of AS in Chinese Han population. The health education of ankylosing spondylitis in a crowd of young peoples who had a manual work shows a great significance to AS for prevention and control. BASFI function index analysis found that drinking water, noise, place of residence associated with sexual with the BASFI function index, the patients drinking tap water and drinking well water in patients compared BASFI poor function index, have a higher noise in the vicinity of the place of residence of the patients compared with patients living in a quieter environment the BASFI poor function index, BASFI function index compared to patients living in urban and non-urban patients living in rural areas, mountainous areas and other poor. Therefore, we believe that moderate drinking water containing minerals, away from the noise, moderate exercise can help improve body function. Gene-environment interaction study found that there was an interaction of alcohol with FCRL4gene, often drinking population popularize the knowledge of the AS and temperance propaganda, has a very important significance for the early stages of the disease prevention and control. |
