Background and purpose: Growing evidences has shown that aberrant changes ingenetics and/or epigenetics may lead to HCC. With recent advances in tumor biology,several reports have suggested that loss of gene function by epigenetic silencingcontributes to HCC. Several study has reported SOX17gene expression was silenced bypromoter region methylation in colon cancer,WNT signaling pathway is an ancient andhighly conserved pathway which determines cell fate and plays crucial roles in bothphysiological and pathophysiological events. The WNTs serve as extracellular signalingmolecules involved in cell differentiation, migration and proliferation during embryonicdevelopment and adult homeostasisSRY-box containing gene17(Sox17) was reportedto be a candidate tumor suppressor and it was an indispensable gene for embryonicdevelopment. Cortactin is cytoskeletal proteinencoded by CTTN gene, It has provedthat CTTN gene is frequently overexpressed in liver cancer. In this sturdy,Weinvestigated the correlation between the expression level of the SOX17and cell’smigration, invasion power of the hepatoma carcinoma cells.Methods:Hepatocytes QSG7701and four forms of hepatoma carcinoma cells LM3、MHCC97L、HepG2、SKHep1were collected. By the method of Real‐time PCR, Trans‐well Assays and scarification, The expression level of gene Sox17as well as the invasion and immigration of these five different cell lines were detected, We adopt the Spearman’s rank correlation to complete the statistical analysis. Results: mRNA’s expression level of SOX17in the hepatocarcinoma cells more than in the l in the compare with the Expression level of Sox17was significantly less than in QSG7701cell lines, but the Expression level of CTTN is Reversed, Expression level of Sox17became increasing in the LM3、HEPG2、MHCC97‐L、SK‐Hep‐1、QSG‐7701cell lines. The average proliferation level in Scarification area in These five cell lines is16.39%,27.78%,43.33%,64.44%,88.06%.decoration of the puncture cells in the trans‐well assay is17.06、38.35、67.44、165.22、187.53respectively. There existed negative correlation between the expression level of Sox17and cells’ immigration in the scarification area, cells’ number on Transwell cabin’ membranes.Conclusion: In tumor cells, the high expression level of Sox17was correlated with less immigration and cells invasion power.
