Objective:To explore the therapeutic efficacy and potential mechanism of casticin (CAS)in the radiation enteritis of mice, It will provide the basic clues and theoraticalfoundations for CAS used in clinical treatment of the radiation enteritis.Methods:75male mice of clean grade were divided into5groups (every group n=15),including normal control group (Control), radiated control group (IR), low-dose CASgroup (5mg/kg)(IR+LD-CAS), high-dose CAS group (20mg/kg)(IR+HD-CAS) andsalicylazosulfapyridine (SASP) group (IR+SASP). Apart from the group Control,other four groups were all accepted disposable whole body radiation of6MV X-rayswith the dose of10Gy. Corresponding drugs was administrated Intragastrically to thetest group of mice1day before radiation,1hour,1,2,3days after radiationrespectively and with once a day. The mice were sacrificed on the4thday, andmeanwhile we should do such preparations for this experiment as recording weightchanges and survival rate of mice in each group, examining the condition of abdomenand intestine by laparotomy, observing stools consistency by intercepting colon,measuring the height of intestinal villi and the degree of mucosal injury in mice ileumby HE staining, detecting the expression of TLR4and NF-kB in ileal mucosa byImmunohistochemistry, analyzing the lever of IL-6、TNF-a in serum by ELISA. Results:The weight of mice decreased significantly after the radiation, CAS couldreduce the weight loss and improve the survival rate of mice; radiation caused seriousintestinal inflammation to mice of the group IR, and there were nearly all loose, liquidand poorly formed stools throughout the colon, while CAS could meliorate thesymptoms and there were solid, brown and well-formed stools in colon; Meanwhile,CAS could reduce villi shedding and repair mucosal injury; Immunohistochemistryindicated that CAS could down-regulate the expression of TLR4, NF-κB in the ilealmucosa of irradiated mice; ELISA showed that CAS could inhibit the release of IL-6、TNF-a in serum.Conclusion:1. CAS could reduce villi shedding, meliorate mucosal injury, decrease thesymptom of laxness, relieve the degree of weight loss and improve survival rate.2. the mechanism of the treatment of radiation enteritis may have relationshipwith inhibiting the Signaling pathway of TLR4/NF-κB and reducing the release ofIL-6、TNF-a.3. The therapeutic effects of the high-dose is better than the lower one, bothof them no obviously adverse reaction.
