| ã€Objective】:To investigate the effect of Feixin decoction(FXD) Drug-containingserum on PHDs-HIF-1α expression of Rat Pulmonary Artery Smooth MuscleCells(PASMCs) upon Exposure to hypoxia in Vitro,to explore the relationship withFeixin decoction containing serum and the proliferation of Rat PASMCs upon exposureto hypoxia.ã€Methods】: The part ofcell proliferation:The tissue adherent and cultured primary ratPASMCs. The generation of rats’ Pulmonary Artery Smooth Muscle Cells used thetissue adherent method,detect cell viability after interference with hypoxia(1%O2,94%N2and5%CO2) of12hour use MTT assay and EdU DNA Proliferation invitro Detection. The part of gene expression:Western-blot used to detect the expressionof HIF-1α with hypoxia at different time points,then chose the best point-in-time as thefollow-up time points. The experiment is designde to four groups: the normoxiagroup,the hypoxia group,the hypoxia+FXD group,the hypoxia+nifedipinegroup;Western-blot used to detect the expression of HIF-1α,PHDs.ã€Results】1.FXD Drug-containing serum can inhibit rats’ Pulmonary Artery Smooth Muscle Cellsproliferation in a dose-dependent manner at hypoxia for12hour.2.Effect of the expression of HIF-1α upon Exposure to hypoxia with different timepoints Western Blot results showed that:With the extension of the hypoxia,HIF-1αprotein expression showed an increasing trend,hypoxia12h group reached a peakvalue,higher than the hypoxia2h group (p<0.05),so did hypoxia24h group.3.Effect of the expression of HIF-1α, PHDs protein upon Exposure to hypoxia for12hour Western Blot results showed that:Compared with the control group, the group with different drug concentrations of the HIF-1α, PHD3protein expression weresignificantly increased (p<0.01).4.Effect of the expression of HIF-1α, PHDs mRNA upon Exposure to hypoxia for12hour by RT-PCR Compared with the control group,the group with different drugconcentrations of the HIF-1α, PHD2, PHD3mRNA were significantly increased(p<0.01).5.Effect of FXD Drug-containing serum on expression of HIF-1α, PHDs protein uponExposure to hypoxia for12hour by Western Blot Compared with the model group,thehypoxia+FXD group’HIF-1α protein expression increased(p<0.05),the expression ofPHD1increased even more(p<0.01).6.Effect of FXD Drug-containing serum on expression of HIF-1α, PHDs mRNA uponExposure to hypoxia for12hour RT-PCR results showed that:Compared with themodel group, the hypoxia+FXD/nifedipine group’PHD1mRNA expressionincreased(p<0.05).ã€Conclusions】1.FXD Drug-containing serum can inhibit rats’ Pulmonary Artery Smooth Muscle Cellsproliferation at hypoxia for12hour;2.FXD Drug-containing serum can inhibit the HIF-1α protein expression in rats’Pulmonary Artery Smooth Muscle Cellsat hypoxia for12hour,but can promote theexpression of PHD protein and mRNA;3.May be FXD Drug-containing serum through PHD-HIF-1α pathway to inhibit theproliferation of rat Pulmonary Artery Smooth Muscle Cells at hypoxia for12hour. |
