Objectives: The purpose of this study was to investigate the expression of miR-181aand miR-181b in different degree of differentiation of human gastric cancer cells andtissues, and to explore their role in the development and progression of gastric cancer,which provided a new theoretical basis for the prevention, diagnosis and treatment ofgastric cancer.Methods: Three different degree of differentiation gastric cancer cells (AGS,SGC-7901, MGC-803) and normal gastric mucosa cells GES-1were cultured in vitro.And28cases of cancerou tissues and normal tissues samples of gastric cancer patientswith surgical removal were collected. Used qRT-PCR method detected the expressionlevels of miR-181a and miR-181b in these cells and tissues. Then compared thedifference of the expressions of miR-181a and miR-181b between the gastric cancercells and normal gastric mucosal cells, the gastric cancer tissues and normal tissues.Results:Detected by qRT-PCR method, we found that the expression levels ofmiR-181a and miR-181b in the AGS, SGC-7901and MGC-803three gastric cancercells were higher than in the GES-1cells (P<0.05). The expression levels of miR-181aand miR-181b between AGS, SGC-7901and MGC-803gastric cancer cells were nosignificant differences (P>0.05). Compared with normal gastric tissues, the expressionof miR-181a and miR-181b in gastric cancer tissues was significantly higher (P<0.05).The expression levels of miR-181a and miR-181b in gastric cancer tissues in stage Ⅲ/Ⅳ were higher than in stage Ⅰ/Ⅱ(P<0.05). And their expression levels in gastriccancer patients with lymphnode metastasis were higher than in patients withoutlymphnode metastasis (P<0.05).There was no significant relationship between theexpression levels of miR-181a, miR-181b and age, gender, degree of tumor differentiation (P>0.05).Conclusions: MiR-181a, miR-181b were high expression in gastric cancer cells andtissues, and their expression levels in gastric cancer were associated with the stage andlymphnode metastasis. they might play the role of oncogenes in the development ofgastric carcinogenesis.
