| Objective: To investigate the infection of HPV in colorectal cancer (CRC) and non-cancerousadjacent tissue(NCAT)and colorectal adenoma by using the gene chip, its clinicopathologicsignificance, explore theHPV infection association with Stat3activity and the expression of IL-17in CRC and NCAT.Methods:95CRC tissues and50NCAT tissues and50colorectal adenoma tissues were allparaffin-embedded tissues. Based on using immunohistochemistry (S-P method) detected theexpression of IL-17and activated p-Stat3in CRC tissues and NCAT tissues before, detecting andgenotyping HPV by the gene chip, analyzing the correlation between the HPV infection and theexpression of activated Stat3in CRC tissues and NCAT tissues, analyzing the correlation betweenthe HPV infection and the expression of IL-17in CRC tissues and NCAT tissues. All statisticalanalysis use the SPSS17.0software.Results:(1) HPV infection rate48.4%in CRC(46/95)was much higher than30%(15/50)inNCAT and28%(14/50)in colorectal adenoma. It was statistically significant comparing amongthem (p<0.05). There is no difference (p>0.05) between the HPV infection in CRC and Clinicalpathology characteristics such as gender, age, differentiation degree, gross type,metastasis oflymph node and TNM staging.(2) HPV16and HPV53were detected in CRC,HPV16was themajor subtype,HPV16infection rate was47.3%(45/95)in CRC and HPV53infection rate was1.1%in CRC. The type of all positive samples in NCAT and colorectal adenoma were HPV16. Allof them were single infection. No multiple infection was dected.(3) There was positivecorrelation(rs=0.377,P=0.000,P<0.05) between HPV infection and Stat3activity in CRC. Nocorrelation (rs=0.056,P=0.700,P>0.05)between HPV infection and Stat3activity in NCAT.(4)There was positive correlation(rs=0.333, P=0.001, P<0.05) between HPV infection and theexpression of IL-17in CRC. No correlation (rs=0.046,P=0.752,P>0.05)between HPV infectionand expression of IL-17in NCAT.Conclusions:(1)There was HPV infection in CRC, and HPV infection in CRC may participate inthe pathogensis of CRC.(2) High risk HPV16was the major subtype in the infection in CRC, nomultiple infection was dected, consistenting with the reported at home and abroad.(3)The HPVinfection was positive correlation with Stat3activity and the expression of IL-17in CRC,Suggesting HPV Infection may through IL-17and Stat3improving the pathogensis of CRC. |
