Tongue squamous carcinoma is one of the common malignant tumors oforal and maxillofacial region at present. The39.95%of the incidence in oralcavity carcinoma is tongue squamous carcinoma. The properties of this cancerare growth-fast,highly malignant,easily lymph node metastasis. Clinictreatment comprehensively used presently are surgery, radiotherapy,chemotherapy etc, but the incidence of postoperative recurrence andmetastasis is higher. It has great clinical value to explore positively effectivepreventive measures and drugs.Scutellarin is a flavonoid extracted from traditional Chinese herb, stemleaf of Scutellaria baicalensis or Erigeron. Studies have confirmed that theflavonoids have pharmacological functions in anti-inflammatory, antimicrobial,anti allergy, anti-tumor and other aspects. The purpose of our study is toexplore the possible antitumor mechanism of scutellarin and providetheoretical and experimental foundation for clinical treatment, throughobserving the effects of scutellarin on the expression of Fas/FasL and cellcycle in human tongue squamous carcinoma Tca-8113cell.Objective:Human tongue squamous carcinoma Tca-8113cells were cultured in vitro.The changes of human tongue squamous carcinoma Tca-8113cell cycles andthe expressions of Fas and FasL were detect to observe the influence ofScutellarin on proliferation and apoptosis of human tongue squamouscarcinoma Tca-8113cell so that to study the anti-tumor effects of Scutellarin.Methods:1.Tongue cancer cells Tca8113which bought in nanjing KaiJibiotechnology limited company were cultured in vitro, used for experiments.2.Experimental groups: The control group, and different concentration of scutellarin groups were set up. The control group was treated with10%FBSin RPMI1640culture medium, while scutellarin groups with80μg/ml,120μg/ml,160μg/ml’s scutellarin cultures of cells according to the result ofpreliminary experiment.3.Flow cytometry was applied to observe the changes of human tonguesquamous cell carcinoma Tca-8113cell cycles.4.Immunocytochemistry methods were applied to observe the effect ofscutellarin on Fas/FasL expression in Tca8113cells.5.Western-blot methods were applied to examine the expression ofFas/FasL in Tca8113cells6.The statistical analysis was performed with the SPSS17.0, usingone-way analysis of variance (ANOVA) and LSD-t test. P <0.05and P <0.01were considered statistically significant.Result:1.The results of Flow cytometry showed: with the increase of scutellarinconcentrations, early DNA synthesis cell after the percentage of G1%increased in different concentrations of scutellarin groups, while DNA S-phasepercentage decreased (S%P<0.05). Comparer with control group, thedifference was statistically significant and this change is associated with thechanges of drug concentration. The160μg/ml concentration of scutellaringroup was best among all scutellarin groups.2.Immunocytochemistry staining results: after Tca8113cells were treatedwith80μg/ml,120μg/ml,160μg/ml scutellarin for48h, the positiveexpressions of Fas and FasL in cytoplasm and cell membrane were shownwith brown color. In the control group, the cell cytoplasm and membrane werestained with very shallow colour, and with the increase of the scutellarinconcentrations, stained colour in the cell cytoplasm and membrane graduallyincreased from pale brown to clear pale brown. The expressions of Fas andFasL were obviously stronger than that of control group. Through statisticalanalysis, the differences were statistically significant among groups (P <0.05).3.Western-Blot analysis showed that the expressions of Fas and FasL in the control group were low. After treatment with scutellarin (80,120and160μg/ml), with the increase of scutellarin concentrations, the expressions ofFas and FasL in the human tongue squamous cell carcinoma Tca-8113cellswere higher than that in the control group. The differences were statisticallysignificant (P<0.05).Conclusion:1.Scutellarin could inhibits the proliferation of tumor cells throughprevention on human tongue squamous carcinoma Tca-8113cells from thestage of G1to S.2.Scutellarin has antitumor functions through enhancement of theexpressions of Fas in human tongue squamous carcinoma Tca-8113cells.
