Cordyceps militaris were used as raw materials, the polysaccharide was extracted with hot water,48mice were gavaged with diffirent doses of polysaccharide, the effects of Cordyceps militaris polysaccharide on the antioxidate activity, immune function and the mRNA expression of spleen cytokines were determined in mice.1. The optimum conditions of the removal of protein in the polysaccharide from Cordyceps militaris, are as follows:pH7, the temperature was60℃, the time was90min, the amount of papain was0.6%. The optimum conditions of decoloration are as follows: decoloration temperature80℃, decoloration time50min, the amount of active carbon1%. Under the condition, the purity percent of polysaccharide was55.4%.2. Compared to the normal control, Cordyceps militaris polysaccharide can increase mice thymus, spleen and liver index, total white blood cells, lymphocytes percentage, monocyte percentage, T lyphocytes percentage and IgG in blood serum and they increased with increasing the concentrations of polysaccharide. At high dose, the thymus index were remarkably significantly increased (P<0.01), the spleen index were significantly increased (P<0.05), the total white blood cells were remarkably significantly increased (P<0.01), T lyphocytes percentage and IgG in blood serum were remarkably significantly increased (P<0.01). Compared with blank control Lymphocytes percentage were significantly increased (P<0.05) in middle dose group and high dose group after treated. Neutrophil percentage was reduced with reducing the concentrations of polysaccharide, and were significantly decreased (P<0.05) at high dose.3. In all dose group, GSH-Px, SOD, CAT and AOC activities were significantly increased(P<0.05) or remarkably significantly increased (P<0.01) compared with normal control. At middle and high dose, MDA levels were significantly (P<0.05) or remarkably significantly (P<0.01) decreased. The results indicated that the Cordyceps militaris polysaccharide could enhance antioxidate activity in mice.4. Compared with normal control the concentration of polysaccharide increasing, the mRNA expression levels of IL-1β、IL-6TNF-a in spleen of mice increased. The mRNA expression levels of TNF-a were significantly increased (P<0.05), the mRNA expression levels of IL-1β and IL-6were remarkably significantly increased (P<0.01). The results indicated that Cordyceps militaris polysaccharide had notable affection on the mRNA expression levels of the cytokines in the spleen of mice.
