Orthogonal tests were designed to explore the optimum condition for WAIDF extracting bychemical methods and enzymatic method. The optimum conditions of extraction technology ofWAIDF were selected by means of chemical methods and enzymatic method. Study on theadsorption capacity of WAIDF for fat,cholesterol and bile acid in vitro. To establish a rat modelwith hyperlipidemia with the method of high fat emulsion for8weeks, i.g. two doses of WAIDF(1.2g·kg-1and2.4g·kg-1) and Xuezhikang (2.4g·kg-1) per day for8weeks’ prevention. The serumlipid level, including the content of TC, TG, LDL-C and HDL-C, and the ability of antioxidation(the activities of MDA, SOD and GSH-Px) were measured. The activities of ALT and AST inserum were tested and the pathological changes were observed to estimate the therapeutic effect ofWAIDF on liver. The level of HL, LPL and LPS was obtained so that we could discuss themechanism of WAIDF reducing the blood lipid in hyperlipidemia rats. To observe the effect ofWAIDF on the rats with hyperlipidemia in regulating serum level of lipid, antioxidation andprotecting liver. Results showed that the optimum conditions of acid treatment were obtained asfollows: radio of liquid-solid1:10, concentration of acid3%, extraction temperature30℃,extraction time80min. Under these conditions extraction rate was up to25.4%,, water-holdingwas12.0and expansibility was6.5, purity coefficient was43%. The optimum conditions of alkalitreatment were obtained as follows: radio of liquid-solid1:10, concentration of alkali10%,extraction temperature30℃, extraction time20min. Under these conditions extraction rate wasup to24.7%,, water-holding was13.36and expansibility was7, purity coefficient was56%. Theoptimum conditions of acid and alkali treatment were obtained as follows: alkali treatment ofradio of liquid-solid1:10, alkali treatment temperature30℃, concentration of alkali10%, acidtreatment of radio of liquid-solid1:10, acid treatment time20min. Under these conditionsextraction rate was up to16.5%, water-holding was33.2and expansibility was10.4, puritycoefficient was79%. The optimum conditions of enzymatic method were obtained as follows:radio of liquid-solid1:20, dosage of diastasic500u, extraction temperature30℃, extraction pH4,dosage of protease0.01g, extraction temperature40℃, extraction pH4. Under these conditionsextraction rate was up to19.7%, water-holding was38.6and expansibility was12,puritycoefficient was88%. Quality of WAIDF was better by enzymatic method than chemical methods.Comparatively speaking,acid and alkali treatment for WAIDF extracting was a economy, useful,lowing cost and to request for instrumentation method. In vitro the adsorption capacity ofWAIDF and pectin for cholesterol,the unsaturated fat and bile acid. the adsorption capacity of pectin was0.993g/g, WAIDF was0.870g/g, the adsorption capacity of saturated fat pectin was0.933g/g, WAIDF was0.747g/g. WAIDF was7.72mg/g and49.89mg/g, was8.75mg/gand53.7mg/g. In the simulated stomach acid conditions the adsorption capacity of pectin andWAIDF for cholesterol were lower than in the intestine pH conditions. The results showed thatWAIDF have certain adsorption capacity on fat, bile acid sodium and cholesterol.WAIDF couldobviously decrease the content of TC, TG, LDL-C and the activities of ALT and AST and increasethe content of HDL-C of serum in hyperlipidemia rats drenched with high fat suspension. WAIDFalso lowered the level of MDA and raised activity of SOD and GSH-Px in liver tissue. The degreeof hepatic steatosis in IDF groups were lighter than those in hyperlipidemia model group,especially the high dose group, which was close to normal essentially. WAIDF played importantroles in enhancing the LPL and HL activity and inhibiting LPS. The results showed that WAIDFhad effect on reducing serum lipid, antioxidation and protecting liver obviously, and they weredose-dependent. It maybe had relationship with the improvement of ability of antiocidation, theactivation of lipid metabolism enzyme activity and the inhibition the generation of fat.
