| Objective:To determine the distribution and expression level of GPC5in prostate cancer tissues group and benign prostatic hyperplasia tissues group, and to evaluate its clinical significance; To evaluate the distribution and expression level of GPC5in prostate cancer cell lines PC3,LNcap and normal prostate cell line RWPE-1.Methods:1.Observation of GPC5protein experssion in prostate cancer and benign prostatic hyperplasia tissues by immunohistochemistry and analyze statistically.2.Observation of GPC5mRNA experssion in prostate cancer and benign prostatic hyperplasia tissues by real-time quantitative PCR and analyze statistically.3.Analyze the relation, Kaplan-Meier survival Multivariate and analysis for overall survival analysis between the expression of GPC5protein and clinicopathologic parameters.4.Observation of GPC5protein experssion in prostate cancer cell lines PC3LNcap and normal prostate cell line RWPE-1.by immunohistochemistry and western blot then analyze statistically.Results:1.Immunohistochemistry showed that in the prostate cancer and benign prostatic hyperplasia tissues, the expression of GPC5protein in prostate cancer tissues was much more lower than the benign prostatic hyperplasia tissues (P<0.01). GPC5protein expression in prostate cancer in age and lymph node had no significant different (P>0.05);but cancer in serum PSA, Gleason score, tumor stage, the expression of GPC5protein showed significant difference (P<0.05).2.Real-time quantitative PCR showed that the quantity of GPC5mRNA in cancer tissues was lower than the benign prostatic hyperplasia tissues (P<0.01). GPC5mRNA expression in prostate in age and lymph node had no significant different (P>0.05); but cancer in serum PSA, Gleason score, tumor stage the expression of GPC5mRNA showed significant difference (P<0.05). 3. GPC5protein expression was no significant association in different group of age, lymph node,while it was significant associated with serum PSA, Gleason score, tumor stage.4. GPC5low-expression was signifantly associated with the poor prognosis of prostate cancer patients.Multivariate Cox proportional hazard model analysis showed that GPC5expression is independent prognostic factor.5. Expression level of GPC5in prostate cancer cell lines PC3,LNcaP was much lower than normal prostate cell line RWPE-1and expression level of GPC5in prostate cancer cell lines PC3was much lower in prostate cancer cell lines LNcaP.Conclusion:1.GPC5lower-expression in prostate cancer tissue was significantly higher than the benign prostatic hyperplasia tissue.2.GPC5low-expression closely associates with the tumorigenesis and progression of prostate cancer.3. GPC5low expression may have some reference value for the diagnosis of prostate cancer, and may become a potential target for treatment of prostate cancer,but also need to be futher studied. |
