| Objectives:To examine the effect of miR-155on angiotensin â…¡ (Ang â…¡)-induced primary mice vascular smooth muscle cell (VSMC) proliferation.Methods:Primary cultured VSMCs from the aorta of C57/B16mice were incubated with Ang â…¡ and miR-155. Cells were counted using CCK-8and Edu and flow cytometric analysis of cell cycle progression was performed. Angiotensin â…¡1type receptor (AT1R) gene and protein expression were measured by real-time PCR and Western blotting.Results:1. Ang â…¡ increased the viability of VSMCs in a dose-and time-dependent manner.2. miR-155opposed the Ang â…¡-induced increase in VSMC viability.3. miR-155inhibited Ang â…¡-induced proliferation of VSMCs.4. miR-155increased the number of VSMCs in Gl phase compared to G2and M cell cycle phases.5. miR-155decreased ATIR gene and protein expression.Conclusion:1. miR-155inhibited Ang@-induced VSMC proliferation.2. miR-155can decreased the AT1R expression. |
