| ObjectiveThis study is about three groups of patients with primary biliary cirrhosis(PBC) and chronic hepatitis B(HBV) and health controls(HC) which were performed inperipheral blood and liver specimens Th17/CD4+T cells ratio and the expression levels of IL-17,Simultaneously analyzed the correlation between the serum levels of IL-17and the markers of liver injuryed gamma-glutamyl transferase (GGT)ã€alkaline phosphatase(ALP),compared with the correlation of peripheral bloodã€liver tissue Th17/CD4+T cells and serum IL-17.With the result of liver biopsy analy the function and clinical significance of Th17/IL-17in PBC patients.Methods1Collect the research object:26cases of PBC patients;20cases of HBV patients and20healthy control group.2The ratio of Thl7cells/CD4+T cell in the peripheral blood and liver tissues were detected by flow cytometry(FCM).3The levels serum of IL-17were measured by enzyme-linked immunosorbent assay(ELISA), compared the correlation between the levels of IL-17and GGTã€ALP.4Compared the correlation of the ratio of Th17/CD4+T cell in the peripheral bloodã€liver tissues and the serum of IL-17.5PBC patients were divided into two groups by liver biopsy results, respectively PBC â… -â…¡ and PBC â…¢-â…£,compared the differences of Th17/CD4+T cell in the peripheral bloodã€liver tissues and the serum of IL-17in PBC patients. Results1PBCã€HBV and HC Th17/CD4+T cells in the peripheral blood levels were (36.53±8.38)ã€(29.85±14.27)ã€(19.51±5.34) respectively, the three groups have statistical significance of difference(P<0.05).PBC patients’ Th17/CD4+T cells in the peripheral blood are higher than that in HBV and HC (P<0.05). HBV patients’ Thl7/CD4+T cells in the peripheral blood are higher than that in HC (P<0.05).2PBCã€HBV and health controls(HC) Th17/CD4+T cells in the liver tissue were (44.44±15.13).(28.62±11.68)%(18.78±5.146) respectively, the three groups have statistical significance of difference(P<0.05).PBC patients’ Th17/CD4+T cells in the pliver tissue are higher than that in HBV and HC (P<0.05). HBV patients’Th17/CD4+T cells in the liver tissue are higher than that in HC (P<0.05).3The serum level of IL-17in the three groups were measured by EL AS A, the serum of level of IL-17in PBCã€HBV and HC were (28.37±5.37)ã€(20.79±4.22)ã€(17.13±4.07) respectively, the three groups have statistical significance of difference(P<0.05). PBC patients’IL-17level in the peripheral blood are higher than that in HBV and HC (P<0.05). PBC patients’IL-17in the peripheral blood are higher than that in HC (P<0.05).4PBCã€HBV and HC ALPã€GGT in the peripheral blood levels were (346.1±216.59)ã€(95.15±37.03)ã€(78.25±19.31) respectively, PBC patients’ALP. GGT level in the peripheral blood are higher than that in HBV and HC (P<0.05). The level of ALPã€GGT in HBV and HC are similar(P>0.05).5The resulted showed PBC patients with the serum level of IL-17and ALPã€GGT were positively correlated (P<0.05). In the peripheralã€liver tissue of Thl7/CD4+T cells and ALPã€GGT are not correlated (P>0.05).6PBC Patients have no correlation between Th17/CD4+T cells and IL-17(p<0.05).7PBC â… -â…¡ and PBC â…¢-â…£ patients’ Th17/CD4+T in the peripheral blood are (31.17±8.3),(38.91±7.45) respectively, The level of Th17/CD4+T in the liver tissue are (34.24±10.35).(48.99±14.89) respectively, They all are different(P<0.05). The level of IL-17in the peripheral blood are (27.05±4.14)ã€(28.77±5.56) respectively, there is no different. ConclusionIn this experiment, we found that PBC patients’ peripheral blood, liver tissue Th17/CD4+T cells and serum IL-17is higher than that of HBV HC. HBV patients’ peripheral blood and liver tissue Th17/CD4+T cells and serum IL-17is higher than that of HC. PBC patients’ IL-17and ALP, GGT are correlated; No correlation between PBC patients’Th17/CD4+T cells in peripheralã€hepatic tissues and serum IL-17, maybe influenced by cytokinesã€chemokines and liver microenvironment. Compared PBC I-II and PBCâ…¢-â…£, the peripheral blood and liver tissue in Th17/CD4+T cells are higher than the former. Th17cells and the secretion of pro-inflammatory cytokines IL-17play an important role in pathogenesis and immune inflammation and fibrosis of liver in PBC. |
