Objective:To measure the levels of HNP1-3、IL-8in the plasma and sputum in patientswith different state of bronchial asthma in acute asthma exacerbation and asthmaremission,and apoptotic rate of peripheral blood polymorphonuclear neutrophils;theexpression of HNP1-3with different severities and grade of bronchial asthma, expressionof IL-8and neutrophil apoptosis, observed the relationship between them;to investigate thepossible role of HNP1-3in bronchial asthma.Methods:Peripheral blood and sputum of sixty patients with acute exacerbation ofasthma,nineteen patients with mild group,twenty-three patients with moderategroup,eighteen patients with severe group, seven patients with mild group aftertreatment,eleven patients with moderate group after treatment,ten patients with severegroup after treatment,and twenty healthy subjects were recuited from June2011toDecember2012, plasma and sputum was separated by centrifugation.levels of HNP1-3andIL-8in it by ELISA, peripheral blood polymorphonuclear neutrophils were isolated byFicol separate.levels of HNP1-3mRNA and Caspase-3by RT-PCR and Western blot.neutrophil apoptosis rate by flow cytometry.Results:1.Levels of HNP1-3in the plasma of each group the levels of HNP1-3in theplasma in acute exacerbation of asthma were higher than healthy controls,in mild groupwere higher than in control group, It was not significant difference between them(P>0.05);it in moderate group and severe group were higher than in controlgroup(P<0.01); clinical remission stage were under than acute exacerbation stage (P<0.01).2.Levels of IL-8in the plasma of each group the Levels of IL-8in the plasma in acuteexacerbation of asthma are proportional to different severities, clinical remission stage wereunder than acute exacerbation stage (P<0.05).3.Cell classified counts of each group neutrophil ratio in blood and sputum in acuteexacerbation of asthma are proportional to different severities, clinical remission stage were under than acute exacerbation stage(P<0.05).4. Levels of HNP1-3mRNA in neutrophils were isolated from blood of each group levelsof HNP1-3mRNA in all of acute exacerbation are increased, It was not significantdifference to between mild group and control group(P>0.05), it in moderate group andsevere group were higher than in mild group and control group (P<0.01), It was notsignificant difference to between moderate group and severe group(P>0.05), clinicalremission stage were under than acute exacerbation stage (P<0.01).5. levels of Caspase-3in neutrophils were isolated from blood of each group It was notsignificant difference to between mild group and control group(P>0.05), it in moderategroup and severe group were decreased than in mild group and control group (P<0.01), Itwas not significant difference to between moderate group and severe group(P>0.05),clinical remission stage were higher than acute exacerbation stage (P<0.05).6. Neutrophil apoptosis rate by flow cytometry of each group acute exacerbation wereunder than control group (P<0.01), moderate group were under than mild group and controlgroup (P<0.01), severe group were under than mild group、moderate group and controlgroup (P<0.01), clinical remission stage were higher than acute exacerbation stage(P<0.01).7. Levels of HNP1-3in the sputum of each group mild group were higher than controlgroup (P<0.01), moderate group and severe group were higher than mild group and controlgroup (P<0.01), It was not significant difference to compare with moderate group andsevere group (P>0.05). clinical remission stage were under than acute exacerbation stage(P<0.01).8. Levels of IL-8in the sputum of each group It was not significant difference to comparebetween the group mild and control(P>0.05). moderate group were higher than controlgroup and mild group (P<0.01), severe group were higher than mild group and controlgroup (P<0.01), but it was not significant difference to compare with moderate group(P>0.05), clinical remission stage were under than acute exacerbation stage (P<0.05).9.Correlation analysis The sputum HNP1-3level was positively correlated with sputumIL-8level,and the percentage of sputum neutrophils (r=0.782,0.796,respectively P<0.01),The plasma HNP1-3level was negatively correlated with FEV1、FEV1/predictedvalues,PEF(r=-0.689,-0.603,-0.643,respectively P P<0.01),The sputum HNP1-3level wasnegatively correlated with FEV1, FEV1/predicted values,PEF and PEF/predictedvalues(r=-0.713,-0.818,-0.638,-0.720,respectively P<0.01).Conclusion:1.HNP1-3may be involved in the pathogenesis of airway inflammationin acute asthma exacerbation,it is related to the percentage of neutrophils and IL-8, Itsmight be one of important pathogenesis of asthma.2. the expression of HNP1-3evaluated different severities of acute asthma exacerbation.3.The expression of HNP1-3in blood and sputum may be a noninvasive marker of severityof acute asthma exacerbation.
