Objective:(1)To study the anti-inflammatory and anti-allergic effects of Qin hao nosedrops.(2)To establish allergic rhinitis(AR) rats and guinea pigs models, investigate thetherapeutic effects of Qin hao nose drops with allergic rhinitis models.(3) Though Qin haonose drops of local irritation test, cilium toxicity test and acute toxicity test, to evaluate thepreliminary safety of Qin hao nose drops. Methods:(1) Through the tests of ear conchinduced by xylene, rats passive skin allergy caused by antiserum, guinea pig nasal mucousmembrane of blood capillary permeability of the experimental animal models, to observethe anti-inflammatory and anti-allergic effects of Qin hao nose drops.(2) Rats and guineapigs were sensitized and intranasally challenged with ovalbumin (OVA) and Al(OH)3toestablish allergic rhinitis models. To determine serum immunoglobulin E (IgE), specificIgE (sIgE) and interleukin4(IL-4) by ELISA; To take nasal mucosa doinghistopathological examination.(3) Rats for7consecutive days,2times/d, compared thenasal mucous membrane irritation and stimulation reaction score, and nasal mucosa tissuefor histologic examination; The drug dropped into rabbits left eye conjunctiva and theright eye instillation with saline, observing the local irritation response after the treatment,and to irritation symptoms scores;(4) Take in vivo toad palate mucosa cilia model, recordcilia for swinging time under light microscope, and observe morphological changes suchas the cilia quantity loss situation, mucosal integrity.(5) Rats for7consecutive days,2times/d, by scanning electron microscopy study cilia on the surface of mucous membraneorganization structure, the influence of the morphology and mucosal cilia shape, number,arrangement, etc. Results:(1) Qin hao nose drops could inhibit the degree of ear conch induced by xylene.(2) Qin hao nose drops high-dose group could significantly reduce thecapillary permeability of guinea pig nasal mucosa.(3) Qin hao nose drops could reduce therat PCA allergic reaction.(4) Qin hao nose drops may improve the nasal symptoms and the nasalmucosa of AR animal model of pathological changes, lowed the levels of serum IgE, sIgE andIL-4.(5) The rabbit eye conjunctiva after a single dose, Qin hao nose drops in the dosegroup of stimulus-response, scoring3.7points per game for a mild stimulant; Other tomedicine group have no obvious irritation.(6) The rat nasal mucosa irritation experimentresults showed Qin hao nose drops three dose groups average scores were between0.41~1.50, for mild irritation; and nasal mucosa pathological examination was normal.(7) The toadpalate cilium motion duration affect the experimental results showed that the blank nosedrops and Qin hao nose drops high-dose group has little effect on the cilia.(8) Qin hao nosedrops on the rat nasal mucosa cilia and form experiment, Qin hao nose drops had noobvious the toxicity of cillia under electron microscope (9) In the acute toxicity test,administered during the observation period were not obvious symptoms of poisoning and drug-induceddeath. Conclusion:(1) Qin hao nose drops has obvious the anti-inflammatory andanti-allergic effects.(2) Qin hao nose drops can inhibit the IL-4synthesis of AR rats andguinea pigs, reduce the serum levels of IgE and sIgE, and improve nasal pathologicalchanges, Qin-hao nose drops has certain effective treatment on AR in rats and guinea pigs.(3) Qin hao nose drops has no obvious nasal mucosa irritation、cilia toxicity and acutetoxicity effects, this drug for clinical was safety.
