NMR Metabonomic Study Of Abnormal Savda Munziq’s Mechanism Of Effect To The Abnormal Savda Syndrome TypeⅡ Diabets Rat

Objective: to study the mechanism of effect of Abnormal Savda Munziq to the abnormalsavda syndrome typeⅡdiabets rat. Methods: Metabolic profiling of rat serum and urinesamples from normal control group (n=9), T2DM Abnormal Savda syndrome controlgroup (n=6), T2DM Abnormal Savda syndrome ASM low dose (n=10), medium dose (n=8)and high dose (n=8) treatment groups had been measured by NMR technology combinedwith pattern recognition,Orthogonal Projections to Latent Structures DiscriminantAnalysis (OPLS-DA) and partial least square-discriminant analysis (PLS-DA) method hadbeen used to find out the variability serum and urine metabolits of abnormal savdasyndrome typeⅡdiabets rat after oral administration of low、medium and high dose ofAbnormal Savda Munziq (ASM) for21days. Results:(1) The data analysis showedthat compared with normal control group, the concentration lavel of very low-densitylipoprotein (VLDL)、 low-density lipoprotein (LDL) and carnitine increased while theconcentration level of lactate and β-glucose have decreased in the serum of abnormalsawda syndrome control group,(P<0.05). Except relatively lower concentration level ofscyllitol in the serum of T2DM abnormal sawda syndrome group, Other different groupof samples occurred similar metabolic changes in serum, Which have higher concentrationlevel of β-glucose、α-Glucose,lower concentration levels of very low-density lipoprotein(VLDL)、low-density lipoprotein(LDL)、amino acids (including1-methylhistidine、glycine、 Phenylalanine、Glutamate、Tyrosine、 leucine、isoleucine、valine) and Citricacid、β-hydroxybutyric acid、Lactate、 creatine、lanine、α-oxoglutaric acid、Carnitine、succinic acid、 pyruvic acid、 creatinine、 malic acid、glycoprotein、 acetate、acetoacetic acid、compared with normal comtrol group have significant differences,(P<0.05).(2)Compared with normal control group, the concentration lavel of β-glucose、 α-glucose、hippuric acid、2-oxoglutaric acid、 creatine、Phenylalanine have increased while theconcentration level of lactate and critic acid have decreased in the urine of abnormalsawda syndrome rat,(P<0.05); Other different group of samples occurred similarmetabolic changes in urine, Which have higher concentration levels of β-glucose andα-glucose, lower concentration levels of Citric acid、 β-hydroxybutyric acid、Lactate、malic acid、hippuric acid、acetoacetic acid、α-oxoglutaric acid、Carnitine、creatine、leucine、isoleucine、alanine、Phenylalanine and glutamate in urine, compared with normal comtrolgroup have significant differences,(P<0.05).(3)After oral administration of ASM, thereduced concentration level of some metabolits including leucine、 isoleucine、Phenylalanine、valine、critic acid、α-oxoglutaric acid、hippuric acid and formic acid inabnormal savda syndrome typeⅡdiabets rat serum and urine have been increased,togetherwith, the increased concentration level of β-glucose and α-Glucose in abnormal savdasyndrome typeⅡdiabets rat serum and urine have been reduced, compared with abnormalsavda syndrome type Ⅱ diabets rats group have significant differences,(P<0.05)Conclusion: ASM has significant effects to the abnormal savda syndrome typeⅡdiabetsrat serum and urine metabolits, specially the high dose of ASM. It`s mechanism of effectmay achieved by readjusting the altered glucose metabolism and amino acid metabolismcaused by abnormal savda syndrome typeⅡdiabets.
