| Objective:To explore the preventive protection effect of1,25-dihydroxyvitaminD3on myocardial fibrosis in rats with type2diabetes mellitus.Methods:Thirty-four maleSD rats were randomly divided into control group with8rats,type2diabetes mellitus(T2DM)group with12rats and intervention group(type2diabetes mellitus rats intervenedwith1,25-dihydroxyvitamin D3) with14rats.After high-glucose and high-lipid diet for6weeks,the intervention group was given1,25-dihydroxyvitamin D3lavage,and T2DMgroup was given peanut oil in the same dose as1,25-dihydroxyvitamin D3used in theintervention group.Control group was fed with normal diet.The plasma glucose,bodyweight, cardiac weight,cardiac weight index, lactic dehydrogenase and creatine jubasewere detected,and morphology of cardiac tissues was observed after6weeks oftreatment,while the expressions of connective tissue growth factor and transforminggrowth factor-beta1in cardiac tissues were observed with immunohistochemistry andRT-PCR in these three roups.Results:The levels of plasma glucose, cardiac weight index,lactic dehydrogenase and creatine jubase were higher,and the body weight,and cardiacweight were lower in T2DM and intervention group than those in control group(P<0.05).The levels of plasma glucose, body weight, lactic dehydrogenase and creatinejubase were lower in intervention group than those in T2DM(P<0.05). Cardiac weightand cardiac weight index had no significant difference between intervention group andT2DM group(P>0.05).The expressions of connective tissue growth factor andtransforming growth factor-beta1were higher in T2DM group than those in the othergroups(P<0.05).Conclusion:1,25-dihydroxyvitamin D3can protect cardiac of diabeticrats by inhibiting the expression levels of connective tissue growth factor andtransforming growth factor-beta1,and restraining myocardial fibrosis. |
