| Objective: Evaluation of miniature pigs autologous periodontal ligament stem cellscombined with guided tissue regeneration and repair of periodontal two types of bonedefects bone.Method: Six Guizhou miniature pigs were divided into3groups randomly, slowerdesktop machine mill in addition to the established the two types periodontal bone defectmodel, bone defect is located between the third premolar and fourth premolars andexposed fourth premolar mesial root. The experimental group two animals were pulled outon the mandibular canine and got periodontal ligament stem cells, defects were repairedwhich ombined periodontal ligament stem cells with hydroxyapatite bioceramic; negativecontrol group of eight defects simple implant Hydroxyapatite ceramics; blank controlgroup eight defects model implant nothing. After12weeks, three groups of animals wereeuthanized and got specimens. Through clinical examination indicators, skull spiral CTscan and three-dimensional reconstruction and HE staining results determine the threegroups periodontal bone tissue into healing.Results: SPSS17.0statistical analysis based on the clinical examination of indicator datais a significant difference (P <0.05), the experimental group healing is better than the othertwo groups; skull spiral CT experimental group showed no significant difference in bonemineral density of the surrounding normal alveolar bone density. Control group defectdefect still visible cavity was not completely healed. Blank group bone defect is stillvisible; eosin staining results of the experimental group, new bone filled with bone defect,normal calcification of bone structure. Control group, new bone has not been filled withbone defects, a very low degree of calcification. The blank group foreseeable lot ofcollagen fibers, the defect is still a large number of vacancies, no significant new boneformation. Conclusions: The PDLSC combine with GTR can effectively improve the miniature pigtwo types of periodontal bone defect healing and periodontal regeneration ability.Application PDLSC joint HA can better repair of periodontal bone defects, it is one ofthe ideal experimental methods of repair small-scale bone defects. |
